虎杖对大鼠急性四氯化碳肝损伤保护作用的量效关系

目的观察不同剂量虎杖煎剂对四氯化碳(CCl4)致大鼠肝损伤的影响。方法将40只大鼠随机分为5组,即正常对照组、模型对照组及虎杖煎剂高、中、低剂量组,分别腹腔注射等容积的生理盐水、生理盐水及虎杖煎剂(0.8g/ml,0.4g/ml,0.2g/ml)。连续6d,每日1次。末次给药后,除正常对照组皮下注射生理盐水外,其他大鼠皮下注射CCl4原液1次,复制大鼠急性肝损伤模型,检测大鼠血清丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)活性,并取肝组织作病理学检查。结果不同剂量的虎杖煎剂均可明显降低CCl4致大鼠血清ALT和AST活性升高,并呈明显的量效关系。虎杖煎剂组大鼠肝组织变性、坏死的程度与模型对照组比较明显减轻。结论虎杖煎剂对CCl4所致大鼠急性肝损伤具有明显的保护作用和量效关系。

Dose-effect Relationship of Protection of Polygoni on Acute Hepatic Injury of Rats Induced by CCl4

Objective To obserwe the decoction effcets of different doses of polygoni on CCl4-induced acute liver injury in rats.Methods 40 Wistar rats were randomly divided into five groups:the normal control group,the model control group,the high,media and low dose treating group of polygoni decoction injected intraperitoneally with different dosage(0.8g/ml ,0.4g/ml and 0.2g/ml) once a day for six days respectly.After the last injection,all animals except those of normal group which were injected with normal saline were injceted with CCl4 stock solution to induce acute hepatic injury.The activity level of ALT and AST were detected,and the hepatic tissue were identified by pathological examination.Results Different doses of polygoni decoction could decrease the contents of ALT and AST,and had obvious dose-effects relationship.Degree of degeneration and necrosis of hepatic tissues in treatment group was obviously lessened in comparison with the model groups.Conclusion Polygoni decoction plays a very important role in protection the acute liver injury and depends on its dosage.
