甲基强的松龙对大鼠脊髓损伤组织中肾上腺髓质素表达的影响

摘    要:目的:观察甲基强的松龙(MP)对大鼠脊髓损伤(SCI)组织中肾上腺髓质素(AM)表达的干预作用.方法:SD大鼠108只随机分为假手术组、MP干预组(MP组)和脊髓损伤组(SCI组),每组36只.假手术组只切除椎板不损伤脊髓,MP组和SCI组采用Allen WD法制作大鼠急性脊髓损伤模型,并于术后1 h经尾静脉分别给予50mg/kg MP和等量生理盐水.于术后2 h、8 h、1 d、2 d、3 d、6 d取骨髓组织(每个时点6只大鼠),HE染色观察脊髓损伤情况,免疫组化染色观察AM的表达,并于术后1 d、3 d、6 d行运动功能评定.结果:HE染色,假手术组各时间点无明显异常,MP组病理变化轻于SCI组.免疫组化染色,假手术组中AM低表达,SCI组AM表达增高并于术后8 h达峰值,以后逐渐降低(P<0.01),MP组AM的表达在术后8 h、1 d、2 d、3 d均高于SCI组,差异有统计学意义(P<0.05).运动功能评分:假手术组1 d、3 d、6 d均高于MP、SCI组,MP组在术后3 d、6 d高于SCI组,P均<0.01.结论:急性脊髓损伤后AM表达代偿性增高并存在动态变化,MP干预可上调AM的表达从而减轻脊髓的继发性损伤.

Abstract:Aim: To observe the expression of adrenomedullin (AM)in acute spinal cord injury(SCI) tissue of rats and the interference effects of methylprednisolone (MP). Methods: A total of 108 SD rats were allocated into sham operation group, MP SCI group, and SCI group, 36 rats in each group.MP SCI group and SCI group were given 50 mg/kg MP and normal saline,respectively. Spinal cord tissue was taken at 2 h, 8 h, 1 d, 2 d, 3 d, 6 d for HE staining and immunohistochemical staining. At 1 d, 3 d, and 6 d after operation, the motor function was evaluated. Results:HE staining showed that the spinal cord tissue of the sham operation group was normal, and the pathological changes of the spinal cord tissue in MP SCI group were milder than those in SCI group. The immunohistochemical staining showed that the expression of AM in sham operation group was lower. The expression of AM in SCI group increased after operation,peaked at 8 h,and decreased afterwards(P<0.01). The expression of AM in the MP SCI group at 8 h,1 d, 2 d and 3 d was higher than that in SCI group(P<0.05). The motor function scores at 1 d, 3 d,6 d in sham operation group were higher than those in MP SCI group and SCI group. The motor function scores at 3 d, 6 d in MP SCI group were higher than those in SCI group(P<0.01). Conclusion: The expression of AM increases compensatorily after SCI. MP can upregulate the expression of AM thus relieve SCI.
