抑癌基因p27对增生性瘢痕成纤维细胞增殖及DNA合成的影响

摘    要:目的:探讨抑癌基因p27对增生性瘢痕的作用。方法:取体外培养的人增生性瘢痕成纤维细胞(HTsFb),将构建的pcDNA3-p27真核表达质粒体外转染HTsFb,经G418筛选获得稳定性表达的细胞克隆,用MTT和^3H-TdR掺入法观察抑癌基因p27对HTsFb增殖及DNA合成的影响。结果:抑癌基因p27对HTsFb增殖及DNA合成均有显著抑制作用。结论:抑癌基因p27可能通过抑制成纤维细胞增殖及DNA合成抑制瘢痕增生。

Abstract:AIM To investigate the effect of tumor suppression gene p27 on the forming of hypertrophic scars. METHODS The eukaryotic expression plasmid pcDNA3-p27 for tumor suppression gene p27 was transduced into cultured human hypertrophic scar fibroblasts in vitro. MTT and 3H-TdR incorporation were applied to measure the effect of p27 on the proliferation and DNA synthesis of hypertrophic scar fibroblasts. RESULTS Tumor suppression gene p27 inhibited the proliferation and DNA synthesis of hypertrophic scar fibroblasts. CONCLUSION Tumor suppression gene p27 may inhibit scar forming by inhibiting the proliferation and DNA synthesis of fibroblasts.
