大鼠肾发育中Wnt4蛋白的表达

摘    要:摘要:目的 探讨大鼠肾发育中 Wnt4 蛋白的表达规律及 Wnt4 蛋白对大鼠肾发育的影响。方法 选取胚龄(E) 18、 20 d 和生后(P) 0、 1、 3、 5 和 7 d 的大鼠肾脏, 应用免疫组织化学技术, 定性分析该时间段内大鼠肾组织中Wnt4 蛋白的表达部位及其变化情况; 应用蛋白印迹检法, 定量分析该时间段内大鼠肾组织中 Wnt4 蛋白表达量的变化。结果 免疫组化结果显示, 从 E 18 d~P 7 d, Wnt4 蛋白在近端小管, 在生肾区的输尿管芽、 逗号小体以及 S 小体的表达较强, 在远端小管的表达较弱, 在肾小体的表达由强到弱。蛋白印迹检测结果显示, Wnt4 蛋白的表达量 E 18d 时开始逐渐降低, P 1 d 时到达低点, 然后开始回升, P 7 d 时再次降低。结论 在大鼠肾发育中, Wnt4 蛋白通过启动经典的 Wnt/β-catenin 信号通路参与调控肾单位的发育, 启动非经典的 Wnt/PCP信号通路参与调控近端小管的延长, 其可能与肾单位的形成以及近端小管的发育密切相关。

Abstract:Abstract: Objective To explore the expression profile of Wnt4 in rat kidney during renal development and its effect onrenal development. Methods Rats with embryonic age of 18 days (E 18 d) , 20 days (E 20 d) as well as postnatal age of 0 day(P 0 d), 1 day (P 1 d), 3 days (P 3 d), 5 days (P 5 d) and 7 days (P 7 d) were selected. Expression levels of Wnt4 in rat kidneyduring renal development were quantified by immunohistochemistry and Western blot in all time points. Results Immuno⁃histochemistry analysis showed that during E 18 d to P 7 d, Wnt4 mainly expressed in proximal tubules, ureteric bud, commashaped bodies and S shaped bodies of nephrogenic zone; the expression in the distal tubule was weak; the expression in renalcorpuscle decreased with time; Western blot analysis showed that the expression of Wnt4 in rat kidney began to decrease fromE 18 d and reached bottom at P 1 d then rise again until P 7 d when it dropped again. Conclusion During renal development,Wnt4 proteins were involved in the development of the nephrogenic zone through regulating canonical Wnt/β-catenin signalingpathway, and was involved in extension of proximal tubules by inducing the non canonical Wnt/PCP signaling pathway. Expressionof Wnt4 protein in rat kidney was closely related to nephron formation and development of proximal tubules.
