BRCA1在散发性乳腺癌干细胞中的表达及意义

摘    要:目的 探讨乳腺癌易感基因1(breast cancer susceptibility gene 1,BRCA1)在散发性乳腺癌干细胞和分化细胞中的表达及意义.方法 选取散发性乳腺浸润性导管癌新鲜标本30例,采用机械分离法将乳腺癌组织块制备成单细胞悬液,通过免疫磁珠两步法从中分离出乳腺癌干细胞(CD44+/CD24-细胞)和分化细胞(CD24+、CD44-、CD24-细胞),应用免疫细胞化学PV6000两步法分别检测两组细胞BRCA1的表达情况.结果 乳腺癌干细胞所占比例平均为2.96%,乳腺癌干细胞BRCA1阴性组与阳性组相比,乳腺癌干细胞在乳腺癌中的比例明显升高,其差异有统计学意义(P<0.01);BRCA1在乳腺癌干细胞和分化细胞的阳性率分别为53.3%(16/30)、83.3%(25/30),差异有显著性(P<0.05).结论 BRCA1能够抑制乳腺癌干细胞的增殖,某些乳腺癌的癌干细胞在增殖分化过程中出现BRCA1的表达.

Abstract:Purpose To investigate the expression and significance of breast cancer susceptibility gene 1(BRCA1) in sporadic invasive ductal carcinoma stem cells and differentiated cells.Methods 30 fresh specimens of breast invasive ductal carcinoma were made into single cell suspensions by mechanical separation,then breast cancer stem cells(CD44+/CD24-cells) and differentiated cells(CD24+ and CD44-,CD24-cells) were separated by immunomagnetic sorting.Immunocytochemical technique was employed to detect the expression of BRCA1 in two groups.Results Breast cancer stem cells accounted for an average of 2.96%,breast cancer stem cells of BRCA1-negative expression group in proportion was higher than the BRCA1-positive expression group,there was a significant difference(P<0.01).The positive expression of BRCA1 were 16(53.3%) and 25(83.3%) out of 30 breast cancer stem cells and differentiated cells respectively,there was a significant difference(P<0.05).Conclusion BRCA1 can inhibit breast cancer stem cell proliferation,some breast cancer stem cells with BRCA1-negative expression appear the expression of BRCA1 in corresponding differentiated cells.
