水通道蛋白4和高迁移率族蛋白1在晚期胃癌中的表达

【目的】 研究晚期胃癌组织中水通道蛋白4(AQP4)和高迁移率族蛋白1(HMGB1)表达,探讨它们表达的临床意义【方法】 选择1999年11月至2005年6月在中山大学肿瘤医院接受FOLFOX或PLF方案化疗的Ⅳ期胃癌患者60例进行研究，胃癌组织病理蜡块切片,采用免疫组化方法检测其中AQP4和HMGB1蛋白表达情况并进行评分，Kaplan-Meier方法分析HMGB1表达与无进展生存时间(PFS)?总生存时间(OS)的关系【结果】 在60例已行FOLFOX或PLF方案治疗的胃癌患者标本中,3例腺癌标本中AQP4 阳性(5%),肿瘤旁相对正常的胃腺体表达全部强阳性(100%)HMGB1在18例标本中表达(30%)阳性,其中13例来自于FOLFOX治疗组(31%,13/42),5例来自于PLF组(31.3%,5/16),两组的阳性率无差异(P = 0.806)生存分析显示阳性与阴性表达的患者无进展生存期和总生存期差异无统计学意义，【结论】AQP4和HMGB1选择性在部分晚期胃癌组织中表达增加,它们在胃癌的发生，发展中的临床意义需进一步扩大样本研究

Expression of Aquaporin-4(AQP4)and High Mobility Group box-1(HMGB1)in Stage IV Gastric Carcinoma

Abstract: 【Objective】 To detect the expression of HMGB1 and AQP4 in Ⅳ stage gastric carcinoma and to explore their clinical significance. 【Methods】 Sixty samples from Ⅳstage gastric carcinoma patients treated with FOLOFOX or PLF plan were selected. The expression of AQP4 and HMGB1 were determined by immunohistochemical analysis. Kaplan-Meier survival analysis was taken for the relation among HMGB1, progression-free survival(PFS) or overall survival(OS). 【Results】 The expression of AQP4 was detected in 3 of 60 gastric carcinoma(5%), which was significantly lower than those in normal gastric mucosa(60/60,100%). The expression of HMGB1 was detected in 18 (30%, 13 from FOLOFOX group and 5 from PLF group )of 60 gastric carcinoma. There was no significance between HMGB1 negative paitents and positive paitents to progression-free survival and overall survival time. 【Conclusions】 AQP4 and HMGB1 selectively expresse in advanced stage gastric carcinoma.
