利开灵对脑出血大鼠脑组织CD14、TLR4 mRNA表达的影响

摘    要:目的:探讨脑出血后大鼠出血局部脑组织CD14、TLR4 mRNA表达的变化规律,并观察利开灵对其影响。方法:采用Rosenberg法改良后建立大鼠脑出血模型,并根据脑出血不同时点(12、24、48、72h),用RT-PCR方法动态观察假手术组、模型组、利开灵组大鼠出血局部脑组织CD14、TLR4 mRNA的表达,并观察利开灵对神经功能缺损体征及出血局部脑组织相应病理改变的影响。结果:模型组各时点CD14、TLR4 mRNA均明显升高,在48h达最高(P<0.01,P<0.05);同时发现利开灵可明显改善脑出血后神经功能缺损体征及出血局部脑组织病理形态损伤,并下调CD14、TLR4 mRNA过高表达。结论:CD14/TLR4信号通路可能参与了脑出血后病理损伤过程,利开灵对出血局部脑组织CD14、TLR4 mRNA表达有较好的调控作用,这可能是利开灵改善脑出血后脑水肿的作用机制之一。

Abstract:Objective:To explore the dynamic changes of mRNA expression of CD14 and TLR4 in the surroundings of the bleeding region after intracerebral hemorrhage in rats,and to research the effect of Likailing(LKL) Decoction.Methods:Rat models of intracerebral hemorrhage were established by improved Rosenberg method.Rats were randomly divided into sham group,model group and LKL treatment group.Observing the dynamic changes on mRNA expression of CD14 and TLR4 in the surroundings of the bleeding region by RT-PCR at 12,24,48,72 hours,and the effect of Likailing Decoction on signs of neurological deficit and pathological morphology changes in the surroundings of the bleeding region.Results:The mRNA expression of CD14 and TLR4 in the surroundings of the bleeding region of model group rats obviously increased and reach the peak at 48h(P<0.01,P<0.05).Likailing Decoction had the effect in lightening the injury degree on signs of neurological deficit and pathological morphology,and could reduce the mRNA expression of CD14 and TLR4.Conclusion:The CD14/TLR4 signal pathway may be involved in the pathological process after intracerebral hemorrhage.Likailing Decoction had regulatory effect on mRNA expression of CD14 and TLR4 in the surroundings of the bleeding region,which may be a part of the action mechanism of improving effect of Likailing Decoction on brain edema after intracerebral hemorrhage.
