璇机化痞膏定性定量方法研究

摘    要:目的 建立璇机化痞膏(乳香、没药、秦艽、关黄柏、大黄等)的定性定量方法.方法 采用薄层色谱法对关黄柏、乳香、大黄、秦艽进行定性鉴别,并采用HPLC法测定秦艽中龙胆苦苷及大黄中芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚,HPLC条件为C18色谱柱,流动相分别为乙腈-0.1%醋酸(9∶91)和乙腈-甲醇-0.1%磷酸溶液(42∶23∶35),检测波长为274 run和430 nm.结果 薄层色谱斑点清晰;龙胆苦苷、芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚分别在0.031 47 ~0.629 4μg、0.019 92 ~0.498 0μg、0.018 82 ~0.4705 μg、0.021 28~0.532 0μg、0.040 4~1.010μg、0.019 34 ~0.483 5μg范围内呈良好的线性关系,r均大于0.999 9,平均回收率为96.2% ~ 99.6%,RSD为1.69% ~3.61%.结论 该方法简便易行,重复性好,可用于璇机化痞膏的质量控制.

Abstract:AIM To establish the qualitative and quantitative methods for Xuanji Huapi Plaster(Olibanum,Myrrha,Gentianae macrophyllae Radix,Phellodendri amurensis Cortex,Rhei Radix et Rhizoma,etc.).METHODS Phellodendri amurensis Cortex,Olibanum,Rhei Radix et Rhizoma and Gentianae macrophyllae Radix were identified by TLC.Gentiopicrin in Gentianae macrophyllae Radix and aloe-emodin,rhein,emodin,chrysophanol,physcion in Rhei Radix et Rhizoma were determined by HPLC.The HPLC was performed on C18 column with the mobile phase of acetonitrile-0.1% acetic acid(9∶ 91) and acetonitrile-methanol-0.1% phosphate acid(42∶23∶ 35),and detection wavelengths were set at 274 nm and 430 nm.RESULTS The TLC spots developed were clear.Gentiopicrin,aloe-emodin,rhein,emodin,chrysophanol and physcion showed good linear relationship in the ranges of 0.031 47-0.629 4 μg、0.019 92-0.498 0 μg、0.018 82-0.470 5 μg、0.021 28-0.532 0 μg、0.040 4-1.01 0 μg、0.019 34-0.483 5 μg(r≥0.999 9),respectively.Their average recoveries were in the range of 96.2%-99.6% with RSDs of 1.69%-3.61%.CONCLUSION The method is simple and reproducible.It can be used for the quality control of Xuanji Huapi Plaster.
