HPLC法测定祛瘀清热颗粒剂中蒽醌类成分

目的 建立祛瘀清热颗粒剂(大黄、桃仁、桂枝、山楂、炙甘草等)中5种蒽醌类成分的同时测定方法.方法 采用高效液相色谱( HPLC)法.Hypersil BDS C18(5μm,4.6mm×250 mm)色谱柱,流动相为甲醇-0.1％磷酸(79:21),体积流量为1.00 mL/min,检测波长为254 nm.结果 芦荟大黄素、大黄酸、大黄素、大黄酚和大黄素甲醚分别在0.45～28.40 μg/mL(r=0.9998)、0.40 ～25.20 μg/mL(r=0.999 8)、0.59～37.40 μg/mL(r =0.999 8)、1.08～68.80 μg/mL(r=0.9998)和0.22～14.00 μg/mL (r =0.999 9)范围内线性关系良好,平均回收率分别为96.11％、99.04％、97.82％、99.67％和98.63％,RSD分别为2.58％、1.75％、1.59％、0.94％和2.07％.结论 建立的HPLC方法快速、简便、准确,能同时检测祛瘀清热颗粒剂中5种蒽醌类成分,可为祛瘀清热颗粒剂的质量控制提供参考.

Determination of anthraquinones components in Quyu Qingre Granules by HPLC

AIM To determine the contents of five anthraquinones components in Quyu Qingre Granules(Rhei Radix et Rhizoma,Persicae Semen,Cinnamomi Ramulus,Crataegi Fructus,Glycyrrhizae Radix et Rhizoma praeparata cum melle,etc.).METHODS HPLC was used with Hypersil BDS C18(5 μm,4.6 mm×250 mm) and methanol-0.1%phosphoric acid(79∶ 21) as the mobile phase.The flow rate was 1.00 mL/min.The detection wavelength was set at 254 nm.RESULTS The linear ranges were 0.45-28.40 μg/mL(r=0.999 8) for aloe emodin,0.40-25.20 μg/mL(r=0.999 8) for rhein,0.59-37.40 μg/mL(r=0.999 8) for emodin,1.08-68.80 μg/mL(r=0.999 8) for chrysophanol and 0.22-14.00 μg/mL(r = 0.999 9) for physcion,respectively.The average recoveries of the five components were 96.11%,99.04%,97.82%,99.67% and 98.63%,respectively.Their RSDs were 2.58%,1.75 %,1.59%,0.94% and 2.07%,respectively.CONCLUSION The established method is rapid,convenient and precise for the determination of five anthraquinones components in Quyu Qingre Granules.This study can provide information for controlling quality of this medicine.
