白香丹胶囊对愤怒情绪模型大鼠海马GABABR和腺苷酸环化酶表达的影响

摘    要:目的检测愤怒情绪模型大鼠海马γ-氨基丁酸B受体两个亚基GABABR1、GABABR2和腺苷酸环化酶(AC)表达的变化,初步探讨白香丹胶囊(芍药苷、香附挥发油和丹皮酚)对肝气逆证的中枢干预机制。方法大鼠随机分为正常组、模型组、白香丹胶囊组(0.2 g/kg)、巴氯芬组(0.008 g/kg),采用"社会隔离结合居住入侵"方法制备愤怒情绪大鼠模型,药物组按相应剂量灌胃给药7 d,每日1次。通过糖水偏好实验、旷场实验和高架十字迷宫实验进行模型评价,用免疫荧光技术检测各组大鼠海马GABABR1、GABABR2和腺苷酸环化酶的表达变化。结果与正常组比较,模型组糖水偏好系数降低,旷场实验得分增高,高架十字迷宫实验进入开放臂次数百分数(OE%)和开放臂停留时间百分数(OT%)降低,海马GABABR1、GABABR2和腺苷酸环化酶表达水平降低。与模型组比较,白香丹胶囊组和巴氯芬组旷场实验得分降低,OE%和OT%值升高,海马GABABR1、GABABR2和腺苷酸环化酶表达水平升高。结论大鼠海马GABABR1、GABABR2和腺苷酸环化酶表达降低可能与愤怒情绪的产生有关,白香丹胶囊平肝理气的中枢机制可能与其恢复海马的GABABR的表达和功能有关。

Abstract:AIM To detect the expressions of GABABR1,GABABR2 and adenylate cyclase(AC) in hippocampus of rats with anger-out emotion,and to explore the intervention mechanism of Baixiangdan Capsule(paeoniflorin,cyperus oil and paeonol) treating liver-qi invasion syndrome.METHODS The Wistar rats were randomly divided into normal group,model group,Baixiangdan Capsule group(0.2 g/kg) and baclofen group(0.008 g/kg).The anger-out emotion rat models were established with a social isolation plus resident intruder stress.The rats were evaluated by sucrose intake,the open field and elevated plus-maze tests.The expressions of GABABR1,GABABR2 and AC in hippocampus were detected by the immunoflourescence.RESULTS Compared with the normal group,the modle group had the lower sucrose intake,the higher total score of open field tests,the lower open arm entry percent(OE%),the lower open arm time percent(OT%) of elevated plus-maze tests and the lower expressions of GABABR1,GABABR2 and AC in hippocampus.Compared with the normal group,Baixiangdan Capsule and baclofen decreased the total score of open field tests,and increased the OE% and OT% of elevated plus-maze tests and the expressions of GABABR1,GABABR2 and AC in hippocampus.CONCLUSION The lower expressions of GABABR1,GABABR2 and AC in hippocampus may have close relationship with anger-out emotion.Increasing the expression and the function of GABABR in hippocampus may be one of the mechanisms for flatting liver-qi or the antianxiety effect of Baixiangdan Capsule.
