摘    要:目的建立不同干燥工艺干姜的超高效液相色谱(UPLC)特征指纹图谱,为干姜的干燥工艺选择提供依据。方法生姜,晒干、晾干、真空干燥和微波干燥干姜的甲醇超声提取液采用Acquity UPLC BEH C18(2.1 mm×100mm,1.7μm)色谱柱;流动相为乙腈-水溶液,梯度洗脱;检测波长280 nm,分析各样品指纹图谱相似度。结果建立了10批不同干燥工艺干姜药材的UPLC特征指纹图谱,标定了15个共有峰,并指认其中6、8、10-姜酚3个色谱峰;10批干姜药材的相似度为0.980~0.997。结论干姜炮制工艺以晾干和60℃真空干燥为宜。

Abstract:AIM To establish a UPLC method of determining the characteristic chromatographic profiles of Zingiberis Rhizoma(Zingiberis Rhizoma recens) for the choice of appropriate drying method.METHODS Methanol extracts made from Zingiberis Rhizoma recens and Zingiberis Rhizoma processed with drying in sunshine,in the shade,vacuum drying and microwave drying were extracted by ultrasonic extraction.The UPLC characteristic chromatographic profiles of their extracts were determined on an acquity UPLC BEH C18 Column(2.1 mm×100 mm,1.7 μm) with the mobile phases of acetonitrile and water in gradient elution mode.The detection wavelength was set at 280 nm.Similarity analysis was carried out on chromatographic pattern.RESULTS The common mode of the UPLC characteristic chromatographic profile was set up with fifteen common peaks in the fingerprints from ten batches of samples,three of which were identified as 6,8,10-gingerol.The similarity between 10 batches of samples was in the range of 0.980-0.997.CONCLUSION According to the similarities of the fingerprints,drying in the shade and vacuum drying at 60 ℃ are recommended as the preferred processing.
