补骨脂中总香豆素的超临界CO2流体萃取工艺研究

摘    要:目的研究超临界CO2流体法萃取补骨脂中总香豆素的最佳工艺条件。方法在对萃取压力、萃取温度、萃取时间、解析压力和解析温度单因素分析的基础上,利用Plackett-Burman实验找出主要的影响因素,并通过Box-Behnken响应面分析法对重要影响因素进行优化,得出最优的工艺条件。结果超临界CO2流体萃取法提取补骨脂中总香豆素的最佳条件为:萃取压力35 MPa,萃取温度62℃,萃取时间3 h,解析压力8 MPa,解析温度60℃。在最佳的提取条件下,总香豆素得率验证值为0.812%,理论值为0.822%,两者的相对误差为1.22%。结论优化所得的超临界CO2流体萃取工艺条件简便易行,预测值与实际值吻合度高,可用于补骨脂中总香豆素的提取。

Abstract:AIM To study the optimum extraction process of total coumarin from Psoralea corylifolia with SFE-CO2.METHODS Extraction pressure,extraction temperature,extraction time,separation pressure,and separation temperature were selected as casual factors based on single-factor experiments.Plackett-Burman experiment design was used to select the main factors.Box-Behnken response surface methodology was used to optimize these main factors.RESULTS The optimum extraction process was as follows: 35 MPa of extraction pressure,62 ℃ of extraction temperature,3 h extraction time,8 MPa of separation pressure and 60 ℃ of separation temperature.Under these conditions,the extraction rate of total coumarin was 0.812%,validation value was 0.822%,and relative error was 1.22%.CONCLUSION The optimum process of SFE-CO2 is simple and convenient,the observed and predicted values are close to each other.It can be used to extract total coumarin from Psoralea corylifolia.
