蛇毒复方制剂对W256癌细胞诱导的骨癌痛大鼠破骨细胞的影响

摘    要:目的观察冰茶栓(蝮蛇毒、冰片、茶叶、桂枝)对骨癌痛大鼠破骨细胞数量和活性的影响。方法将40只雌性SD大鼠分为冰茶栓组、阳性药对照组、模型对照组及假手术组,采用骨髓腔注射W256癌细胞复制骨癌痛模型;自模型复制第13天开始,冰茶栓组给予冰茶栓101 mg/kg;阳性药对照组给予伊班膦酸注射液20μg/kg,模型对照组和假手术组给予空白栓,末次给药后,取各组大鼠患肢骨组织采用TRAP染色法计数破骨细胞数量、透射电镜观察破骨细胞结构、免疫组化法观察骨保护素表达。结果冰茶栓可明显降低W256细胞诱导的骨癌痛大鼠骨组织破骨细胞数量(P<0.01);诱导破骨细胞凋亡;明显上调骨组织骨保护素表达(P<0.01)。结论冰茶栓可抑制骨癌痛大鼠骨组织破骨细胞的数量和活性。

Abstract:AIM To study the effect of Bingcha Suppository(Agkistrodon halyx Venom,Borneolum,Camelliae Folium,Cinnamomi Ramulus) on the number and activity of osteoclasts in the rat model for bone cancer pain.METHODS Forty female Sprague-Dawley rats were divided into four groups: Bingcha Suppository group,Bondronat group,model group and sham-operation group.The model for bone cancer pain was induced by injecting W256 tumor cells into the rat marrow cavity.The thirteenth day after operation,rats in Bingcha Suppository group were treated with Bingcha Suppository(101 mg/kg),rats in Bondronat group were treated with Bondronat Injection(20 μg/kg),and rats in sham-operation group and model group were treated with blank suppository.At the last administration,osteoclasts in the bone tissues of wound hindlimbs of all rats were counted with the tartrate-resistant acid phosphatase staining.Osteoclasts’ morphology was observed with transmission electron microscopic,and the osteoprotegerin expression was examined by immunohistochemistry.RESULTS Bingcha Suppository could markedly reduce the number of osteoclasts in the bone tissues(P<0.01).It could induce the apoptosis of osteoclasts,and remarkably upregulate the osteoprotegerin expression of the bone tissue(P<0.01).CONCLUSIONBingcha Suppository has a good effect on downregulating osteoclast’s number and activity in rats with bone cancer pain.
