高效液相色谱法测定复方昆丹胶囊中6种主要成分

摘    要:目的建立昆丹胶囊(白芍,佛手,制首乌,白术等)中苯甲酰芍药苷,5,7-二甲氧基香豆素,白术内酯Ⅲ,大黄素,白术内酯Ⅰ,大黄素甲醚的定量测定方法。方法采用Agilent TC-C18柱(250 mm×4.6 mm,5μm),以乙腈-水为流动相进行梯度洗脱,检测波长220nm;柱温30℃;体积流量1.0 mL/min。结果苯甲酰芍药苷、5,7-二甲氧基香豆素、白术内酯Ⅲ、大黄素、白术内酯Ⅰ及大黄素甲醚质量浓度分别在2.20~22.0μg/mL,1.13~11.3μg/mL,1.52~15.2μg/mL,0.95~9.5μg/mL,0.75~7.5μg/mL,0.35~3.5μg/mL(r>0.999 9)范围内线性关系良好。低、中、高3个质量浓度的平均加样回收率分别为97.9%~103.3%(RSD为0.3%~2.4%),99.0%~104.7%(RSD为0.3%~1.6%),95.2%~100.9%(RSD为0.1%~1.7%)。结论方法简单,准确,灵敏,能有效测定昆丹胶囊中多种成分。

Abstract:AIM To establish an HPLC method for simultaneously determining benzoylpaeoniflorin,5,7-dimethoxycoumarin,butenolide Ⅲ,emodin,butenolideⅠand physcion in Compound Kundan Capsule(Paeoniae Radix alba,Citri sarcodactylis Fructus,Polygoni multiflori Radix Praeparata,Atractylodis macrocephalae Rhizoma,etc.).METHODS The analysis was carried out on an Agilent TC-C18 column(250 mm×4.6 mm,5 μm).The mobile phase was composed of acetonitrile(A)-water(B) in gradient elution mode.The detection wavelength was set at 220 nm with the column temperature of 30 ℃.The flow rate was 1.0 mL/min.RESULTS The calibration curves of benzoylpaeoniflorin,5,7-dimethoxycoumarin,butenolideⅢ,emodin,butenolideⅠand physcion were linear in the ranges of 2.20-22.0 μg/mL,1.13-11.3 μg/mL,1.52-15.2 μg/mL,0.95-9.5 μg/mL,0.75-7.5 μg/mL,0.35-3.5 μg/mL(r>0.999 9),respectively.The average recoveries of low,middle and high concentrations of the six components were 97.9%-103.3%(RSD of 0.3%-2.4%),99.0%-104.7%(RSD of 0.3%-1.6%),95.2%-100.9%(RSD of 0.1%-1.7%),respectively.CONCLUSION The method is simple,accurate and sensitive,and it can be used to determine the six components in the preparation.
