宣导泻肺饮对心力衰竭大鼠血流动力学及神经内分泌的影响

摘    要:目的研究宣导泻肺饮对心力衰竭(心衰)大鼠血流动力学及血浆心钠素(ANP)、醛固酮(ALD)、血管紧张素Ⅱ(AngⅡ)的影响。方法 60只Wistar雄性大鼠随机分为假手术组、模型组、中药大剂量组、中药小剂量组、贝那普利(洛汀新)组、中药+洛汀新组。各造模组采用腹主动脉缩窄(结扎)法复制大鼠心衰模型,电磁流量计法和左心室插管法测定血流动力学指标,放免法测定血浆ANP、ALD、AngⅡ浓度。结果与模型组相比,中药大剂量组、中药小剂量组、洛汀新组、中药+洛汀新组心率减慢明显,血浆ANP、ALD、AngⅡ均有不同程度下降,其中中药+洛汀新组在左室收缩压(LVSP)、左室内压最大上升速率(LV+dp/dtmax)、每搏输出量(SV)、心输出量(CO)、每搏量指数(SVI)、心脏指数(CI)等指标改善较其他处理组更加明显(P〈0.05或P〈0.01),中药大剂量组比中药小剂量组在DBP、MAP、LVSP、LV+dp/dtmax、SV、CO、SVI、CI等指标上改善明显(P〈0.05或P〈0.01)。结论腹主动脉缩窄(结扎)法所致心衰大鼠心功能下降,血浆ANP、ALD、AngⅡ分泌增加;宣导泻肺饮可以改善心功能并抑制血浆ANP、ALD、AngⅡ过度分泌。

Abstract:Objective To study the effect of Xuandaoxiefeiyin(XDXFY) on hemodynamics,plasma atrial natriuretic peptide(ANP),aldosterone(ALD),and angiotensin Ⅱ(Ang Ⅱ) in heart failure rats.Methods Sixty-five male Wistar rats were randomly divided into six groups:Sham-operated group,the model group,high-dose XDXFY group,low-dose XDXFY group,Lotensin group,and XDXFY+Lotensin group.The rat models with heart failure were established by abdominal aorta ligation.The hemodynamics was studied by left ventricular catheterization and electromagnetic flow meter.ANP was tested by RIA assay.Results Compared with the model group,the HR slowed down significantly in the high-dose XDXFY group,low-dose XDXFY group,Lotensin group,and XDXFY+Lotensin group.Other indicators were increased(P0.05 or P0.01).Plasma ANP,ALD,and Ang Ⅱ decreased.LVSP,+dp/dtmax,SV,CO,SI,CI in XDXFY+Lotensin group improved significantly(P0.05 or P0.01).The DBP,MAP,LVSP,+dp/dtmax,SV,CO,SI,and CI in high-dose XD group improved significantly than that in low-dose group(P0.05 or P0.01).Conclusion Xuandaoxiefeiyin can improve heart function and inhibit excessive secretion of plasma ANP,ALD,and AngII.
