二氮嗪预处理骨髓源性心肌干细胞移植治疗心肌梗死的实验研究

摘    要:目的探讨二氮嗪预处理骨髓源性心肌干细胞(MCSCs)移植治疗心肌梗死的机制及效果。方法通过单细胞克隆培养技术,从雄性幼鼠骨髓间充质干细胞中筛选具有心肌特异分化潜能的MCSCs;将雌性大鼠心肌梗死模型随机分为对照组、MCSCs移植组、二氮嗪预处理MCSCs移植组;细胞移植后4周,用M型超声心动图检测心功能恢复情况,心脏组织切片进行Masson染色,利用图像分析检测胶原纤维比率,用原位荧光杂交法标记心肌组织中Y染色体细胞,并检测其cTnT的表达。结果二氮嗪预处理细胞移植组与MCSCs移植组相比,大鼠的心功能指数明显提高,胶原纤维比率明显减小,Y染色体阳性细胞数明显增多,差异均有统计学意义。结论二氮嗪预处理MCSCs移植治疗心肌梗死,有利于移植细胞在受损心肌内的存活,并促进心功能恢复,其疗效优于单纯干细胞移植。

Study on Transplantation of Marrow-derived Cardiac Stem Cells Pretreated with Diazoxide for Treatment of Myocardial Infarction

Abstract:Objective To explore the effect of transplantation of marrow-derived cardiac stem cells(MCSCs) pretreated with diazoxide on cardiac function after myocardial infarction(MI).Methods MCSCs were selected from marrow mesenchymal stem cells of male rats with single-cell cloning culture.Thirty minutes after the establishment of acute myocardial infarction model of female rats,the rats were injected at the edge of infracted area with PBS,MCSCs,MCSCs pretreated with diazoxide respectively.Four weeks after MI,the cardiac function was detected by M-mode echocardiography.The infarct size and capillary density were evaluated by Masson and immuno-staining respectively.The donor cells in myocardial tissue were marked with Y chromosome through Fluorescence in situ hybridization.Results Functional recovery of rat hearts,myocardial scar size reduction and the Y chromosome-positive cells of the MCSCs pretreated with diazoxide transplantation group were superior to MCSCs transplant group,and the difference was significant.Conclusion MCSCs pretreated with Diazoxide could improve the survival rate of the transplanted cells and attenuates cardiac injury of MI rats.
