| 摘 要： | 目的丙型肝炎病毒NS5B基因转染人肝癌细胞系Hep3B细胞,检测NS5B的表达与细胞内RNA聚合酶活性。方法使用脂质体把包含全长丙型肝炎病毒NS5B基因的重组载体pcDNA-5B转染到Hep3B细胞。RT-PCR和Wes-tern blot鉴定NS5B基因的表达,荧光素酶检测NS5B的细胞内RNA依赖的RNA聚合酶活性。结果RT-PCR和Westernblot发现转染NS5B基因的Hep3B细胞产生NS5B mRNA和NS5B蛋白,荧光素酶分析表明Hep3B细胞表达的NS5B蛋白具有细胞内RNA依赖的RNA聚合酶活性。结论人肝癌细胞系Hep3B细胞可以成功表达NS5B基因,且表达的NS5B蛋白具备细胞内RNA依赖的RNA聚合酶活性。
