Abstract:In order to further explore the synaptic regulatory mechanism of the hypothalamic neuroendocrine, the multiple innervations of neurons in the hypothalamic paraventricular nucleus(PVN)of rats have been studied by combined method of electron microscopic cytochemistry with double immunoelectron microscopy.First, the CA nerve terminals were lesiond by 6-OHDA,then the peptidergic(OT or SP)and GABAergic neurons were double labeled by pre embedding immunoelectron microscopy using DAB and TMB as chromogens. The results showed that there were peptide(OT,SP)and amino acid(GABA)-like neurons and CA nerve terminals in the PVN,OT neurons received GABA and CA innervations through axo-dendritic synapses,SP neurons received CA and SP innervations through axo-dendritic synapses and the GABA neurons might receive OT,SP,CA and GABA innervations through axo-dendritic synapses. The present study provided the ultrastructural evidences for the multiple innervations of the hypothalamic neurons for the first time. Its significance lies in providing the morphological basis for simultaneously investigating the multiple synaptic regulations of the hypothalamic neuroendocrine.
