C-反应蛋白抑制活化的巨噬细胞分泌基质金属蛋白酶

摘    要:目的研究C-反应蛋白(CRP)对巨噬细胞分泌基质金属蛋白酶(MMPs)的影响。方法以不同浓度的CRP和/或50μg/ml氧化的低密度脂蛋白(ox-LDL)处理THP-1巨噬细胞,应用明胶酶谱法测定细胞培养上清液中MMP-2、MMP-9、MMP-1的活性。结果THP-1巨噬细胞在基础状态下分泌MMP-2、MMP-9、MMP-1,CRP抑制其分泌,以MMP-2为例,CRP抑制MMP-2的酶原(0.82±0.051,与空白对照组比较P<0.01)和其活性形式(0.61±0.030,与空白对照组比较P<0.01)。ox-LDL诱导MMPs的分泌,但这种作用被同时存在的CRP所抑制(0.56±0.098,与ox-LDL组比较P<0.01)。结论CRP抑制活化的巨噬细胞分泌MMPs,并抑制ox-LDL对MMPs的诱导作用。

Abstract:Objective To clarify the effects of CRP on the secretion of MMPs(matrix metalloproteinase) by macrophage.Methods THP-1 macrophages were treated by CRP of different concentrations and/or ox-LDL(oxidized lowdensity lipoprotein) of 50μg/ml,MMP-2,MMP-9 and MMP-1 in culture medium were measured by gelatin SDS-PAGE zymography.Results MMP-2,MMP-9,MMP-1 were secreted by THP-1 macrophages in control group,and were decreased by CRP.For example,Pro-MMP-2(0.82±0.051 VS control group,P<0.01)and MMP-2(0.61±0.030 VS control group,P<0.01)were decreased significantly by CRP.Furthermore CRP inhibited the induction of MMPs by oxLDL(0.56±0.098 VS ox-LDL group,P<0.01).Conclusion CRP inhibited the secretion of MMP-2,MMP-9,MMP-1 by macrophage,and also inhibited the induction of MMPs by ox-LDL.
