艾塞那肽预处理对过氧化氢损伤的H9c2心肌细胞的保护作用研究

摘    要:目的:研究胰高血糖素样肽1(GLP-1)类似物艾塞那肽(EX)预处理对过氧化氢损伤的H9c2心肌细胞的保护作用。方法:取体外培养的H9c2心肌细胞,分为正常对照组、模型组和0·01、0·1、1、10nmol·L-1EX预处理组。除正常对照组外,其余各组用过氧化氢诱导H9c2心肌细胞建立氧化应激损伤模型,EX预处理组建模前30min加入相应浓度EX,共培养6·5h后,检测各组细胞细胞活力和培养液上清中乳酸脱氢酶(LDH)、超氧化物歧化酶(SOD)、肌酸激酶同工酶(CK-MB)活性和丙二醛(MDA)含量。结果:与正常对照组比较,模型组H9c2细胞活力、SOD活性明显降低,LDH、CK-MB活性和MDA含量明显升高(P<0·05)。与模型组比较,0·1、1、10nmol·L-1EX预处理组细胞活力、SOD活性明显升高,LDH、CK-MB活性和MDA含量明显降低(P<0·05),并与剂量成正相关。结论:EX预处理对H9c2心肌细胞的氧化应激损伤有保护作用,并与剂量呈正相关。

Abstract:OBJECTIVE:To study the protective effects of GLP-1 analogue exenatide(EX) pretreatment on H9c2 cardiomyocytes after oxidative stress injured by H2O2.METHODS:Cultured H9c2 cardiomyocytes in vitro were divided into normal control group,model group and EX pretreatment groups(0.01,0.1,1,10 nmol·L-1).Except for normal control group,oxidative stress injury model of H9c2 cardiomyocytes was induced by H2O2 in those groups.H9c2 cells were pretreated with different doses of EX 30 min before modeling for 6.5 h,then cell viability,the activity of LDH,SOD,CK-MB and the content of MDA were determined.RESULTS:Compared with normal control group,the viability of H9c2 cells and SOD were decreased in model group and the activity of LDH,CK-MB and the content of MDA were increased significantly(P<0.05).Compared with model group,the viability of H9c2 cells and SOD were increased significantly in EX pretreatment groups(0.1,1,10 nmol·L-1) and the activity of LDH,CK-MB and the content of MDA were decreased significantly(P<0.05),which was positively related with drug dosage.CONCLUSION:Exenatide pretreatment can effectively protect H9c2 cardiomyocytes from oxidative injury with a concentration-dependent manner.
