外阴鳞状细胞癌组织中VEGF和Bcl-2基因组蛋白H3K4三甲基化水平的表达

摘    要:目的:检测外阴鳞状细胞癌患者癌组织血管内皮生长因子(VEGF)和Bcl-2基因组蛋白H3K4三甲基化(me3)甲基状态及其mRNA的表达。方法:采用染色质免疫共沉淀一实时定量聚合酶链反应(ChIP—qPCR)技术对15例外阴鳞状细胞癌患者癌组织和毗邻正常组织VEGF和Bcl-2基因组蛋白H3K4me3甲基化状态进行定量分析;随后采用定量反转录聚合酶链反应(qRT—PCR)检测基因的mRNA表达。结果:外阴鳞状细胞癌组织中VEGF和Bcl-2基因呈现组蛋白H3K4me3水平增高(P〈0.05):VEGF和Bcl-2基因的mRNA表达增加。结论:外阴鳞状细胞癌组织VEGF和Bcl-2基因可能参与外阴鳞状细胞癌的发生,成为外阴鳞状细胞癌新的治疗靶点。

The expression of histone H3 lysine 4 trimethylation of VEGF and BCL - 2 gene in vulvar squamous cell carcinoma tissue

Abstract:Objective: To detect the expression of the histone H3 lysine 4 trimethylation (H3K4me3) of the VEGF and Bcl- 2 gene in vulvar squamous cell carcinoma (VSCC). Methods: Chromatin immuno- precipitation - quantitative PCR was used to detect the methylation level of VEGF and Bcl - 2 gene in 15 patients with VSCC and the control tissues. The mRNA expression on the VEGF and Bcl - 2 gene was performed by qRT - PCR. Results: Compared to normal tissues, The H3K4me3 of VEGF and Bcl - 2 gene in VSCC increased ( P 〈 0.05). The expression of mRNA of VEGF and Bcl - 2 gene also increased. Conclusions: The VEGF and Bcl - 2 gene may be in- volved in the pathogenesis of VSCC, which may become a novel potential biomarker for the treatment of VSCC.
