紫外分光光度法测定市售檀香中总黄酮含量

摘    要:目的:建立一种可操作性强的市售檀香总黄酮含量测定方法。方法:以芦丁为对照品,利用紫外分光光度法测定檀香中的总黄酮含量。结果:芦丁标准品和檀香分别经过AlCl3-NaOH-NaNO2显色后最大吸收波长为508 nm,其浓度在9.17~55.0μg/mL范围内与吸光度线性关系良好,回归方程为:A=11.714·C+0.0503(R2=0.9995,n=5),平均回收率为98.11%(RSD=1.03%)。市售檀香总黄酮平均含量为2.88 mg/g。结论:本方法简便易行,结果准确,重复性好,可作为檀香总黄酮含量测定的一种手段,用于檀香的日常分析及质量控制。

Abstract:Objective To establish an operable method for the determination of total flavonoids in sandalwood. Methods The content of total flavonoids in sandalwood was determined by Ultraviolet Spectrophotometry with rutin as control. Results The maximum absorption wavelength of rutin standard and sandalwood was 508 nm after coloration with AlCl3-NaOH-NaNO2. The linear relationship between the concentration and absorbance was good in the range of 9.17~55.0 μg/mL. The regression equation was A=11.714.C+0.0503(R2=0.9995, n=5), and the average recovery was 98.11%(RSD=1.03%). The average content of total flavonoids in sandalwood was 2.88 mg/g. Conclusion The method is simple, accurate and reproducible. It can be used as a method for determination of total flavonoids in sandalwood and for daily analysis and quality control of sandalwood.
