丹参注射液对胃黏膜Na+-K+-ATPase活性和胃黏膜屏障功能的影响

摘    要:目的探讨丹参注射液对严重腹腔感染状态下大鼠胃黏膜Na -K -ATPase活性变化和电位差的影响。方法将180只大鼠随机分为感染组(盲肠穿孔)、丹参组(盲肠穿孔感染后注射复方丹参注射液)和对照组,每组60只。用紫外分光光度计检测穿孔前和穿孔后3、6、12、24和48h各时相组大鼠胃黏膜Na -K -ATPase活性。应用电生理记录仪检测穿孔前和穿孔后3、6、12、24和48h胃黏膜电位差(GTPD)的变化。结果感染组在盲肠穿孔后3hNa -K -ATPase活性显著降低(P<0.05),12h降至最低(P<0.01),48h仍未恢复正常;丹参组12h、24hNa -K -ATP酶活性比感染组显著升高(P<0.05)。GTPD穿孔后3h即有下降,6h显著下降(P<0.05),12h下降至最低(P<0.01),24h和48h仍然显著低于对照组(P<0.05);丹参组12h、24h比感染组明显升高(P<0.05)。结论严重腹腔感染状态下胃黏膜Na -K -ATPase活性降低,可能是引起胃黏膜屏障功能受损的主要原因,提示早期应用丹参能有效地预防应激性溃疡的发生。

Abstract:Objective To investigate the effects of Dansen on gastric mucosal Na K ATPase activity and gastric transmucosal potential difference during severe intraperitoneal infection in rats. Methods The intraperitoneal infection rat model was established by cecal ligation and puncture(CLP). Assay of Na K ATPase activity in gastric mucosal tissue was conducted by biochemistry method. The electric physiological recorder meter was used to measure gastric mucosal potential difference. Results The activity of Na K ATPase markedly decreased in infection group at 3 h after perforation, compared with the control group(P< 0.05). There was a minimum of Na K ATPase activity at 12 h after perforation in infection group (P< 0.01), was only 48.5% of that in the control group. At 12 h and 24 h post perforatively, Na K ATPase activity of Dansen group significantly increased as compared with infection group(P< 0.05). Gastric transmucosal potential difference (GTPD)in infection group decreased significantly as compared with the control group (P< 0.05) at 6 h after perforation, and rapidly dropped to the lowest at 12 h(P< 0.01). At 12 h and 24 h after perforation , GTPDs were higher in Dansen group than those in infection group(P< 0.05, P< 0.05). Conclusion The reduction of Na K ATPase activity may play an important role in gastric mucosal barrier damage following severe abdominal infection induced by CLP,and the application of Dansen in earlier period can prevent the occurrence of stress ulcer.
