逆转录病毒介导的人卵巢癌 p53基因治疗体外及小鼠体内实验研究

摘    要:目的 :评价人卵巢癌的 p5 3基因治疗效果。 方法 :用携带人野生型 p5 3的逆转录病毒转导 p5 3蛋白表达缺如的人卵巢癌细胞系SK OV 3,通过体外及小鼠体内实验研究转基因的表达及对肿瘤的抑制作用。结果 :逆转录病毒介导的 p5 3基因能够在体外及小鼠体内人卵巢癌细胞SK OV 3中表达 ,并对人卵巢癌生长有抑制作用。体外抑制率为 5 7%~ 88% ;对体内肿瘤抑制率为 40 %。结论 :逆转录病毒介导的外源野生型p5 3能够抑制人卵巢癌生长 ,增加病毒滴度将进一步提高体内治疗效果

Abstract:Objective: To estimate the efficiency of gene therapy with p53 for human ovarian cancer. Methods: After transducing retrovirus mediated p53 gene to human ovarian carcinoma cell line SK OV 3, gene expression and tumor inhibition were studied in vitro and in vivo . Results: Retrovirus mediated p53 could express in SK OV 3 and inhibit tumor growth. The tumor cells were reduced 57% 88%( in vitro ) and 40% ( in vivo ). Conclusion: Exogenous wild type p53 mediated by retrovirus could inhibit the growth of ovarian carcinoma. Increasing titers of virus would make the therapy more effective in vivo .
