噬菌体抗体库技术的发展及未来

摘    要:噬菌体表面展示技术是近年发展起来的 ,将外源蛋白基因与噬菌体表面特定蛋白基因在其表面融合表达的新技术。应用PCR技术从生物体内扩增出整套抗体轻、重链可变区基因 ,克隆到λ噬菌体表达系统 ,构建噬菌体抗体库 ,通过抗原亲和吸附 -洗脱 -增殖 ,从中筛选富集多种目的克隆 ,制备相应单克隆抗体。

Abstract:The exterior protein gene and the specific protein gene coded on the surface of phage can be fusedly expressed on the surface of phage with the recently developed technology-phage display technique. Through the PCR technique, the whole light chain genes and heavy chain genes can be amplified. Cloning these genes into lambda phage expression vectors and constructing phage antibody library through antigen adsorption-elution-proliferation, many targeting clones could be selected and the responding mono-clone antibody could be produced. Detailed progress in this regard was reviewed in the paper.
