脂联素对高糖环境下人视网膜色素上皮细胞分泌VEGF及PEDF的影响

摘    要:目的研究脂联素对高糖环境下人视网膜色素上皮(hRPE)细胞分泌血管内皮生长因子(VEGF)和色素上皮衍生因子(PEDF)的影响。方法hRPE细胞分为3组:①正常对照组;②高糖组;③高糖+不同浓度的脂联素组(2.5,5,10,20μg/ml),分别在48h后,用ELISA法测定上清液中VEGF及PEDF的含量。结果与正常对照组相比,高糖组VEGF水平明显升高,PEDF水平明显下降,加入2.5μg/ml脂联素对VEGF及PEDF水平影响不大,脂联素浓度达到5~20μg/ml时和高糖组相比,结果相反,VEGF水平显著降低、PEDF水平显著升高,差异有统计学意义(P〈0.01)。结论5~20μg/ml脂联素可以使hRPE细胞VEGF分泌减少和PEDF分泌增多。提示脂联素可以纠正高糖诱导的hRPE细胞功能紊乱,通过调节VEGF与PEDF的平衡来抑制糖尿病视网膜病变(DR)中病理性新生血管的形成。

Abstract:Objective To investigate the effects of adiponectin on the expression of VEGF and PEDF of hRPE ceils cultured with high glucose in vitro. Methods The hRPE cells were divided into normal glucose group , high glucose group, high glucose plus adiponectin group(2. 5, 5, 10, 20 μg/ml). 48 hours later, the level of VEGF and PEDF was measured by enzyme-linked immunosorbent assay ( ELISA). Results Compared with normal glucose group , the VEGF level significantly increased and PEDF significantly decreased in high glucose group. Compared with high glucose group , the VEGF level significantly decreased and PEDF significantly increased in high glucose plus adiponectin group (5 -20 μg/ml). (P 〈0. 01 ). Conclusion 5 -20 μg/ml adiponectin can decrease VEGF and increase PEDF of hRPE. Adiponectin can correct the hRPE cell dysfunction induced by high glucose and regulate the balance of VEGF and PEDF to inhibit the formation of new blood vessels in diabetic retinopathy (DR).
