微小RNA-454、微小RNA-132、液泡膜-ATP酶表达与结肠癌临床病理特征关联性分析

摘    要:目的 探讨组织微小RNA(miR)-454、miR-132、液泡膜-ATP酶(V-ATPase)表达与结肠癌病人临床病理特征关联性。方法 选取2011年9月至2014年11月新乡医学院第一附属医院行手术切除的96例结肠癌组织标本、96例癌旁正常结肠组织标本、82例良性结肠病变标本为研究对象。以实时荧光定量逆转录聚合酶链反应(qRT-PCR)测定标本中miR-454、miR-132、V-ATPase mRNA表达,分析miR-454、miR-132、V-ATPase mRNA表达与结肠癌病人一般资料、临床病理参数的关系,随访5年,统计miR-454、miR-132、V-ATPase高表达、低表达病人3年、5年生存率。结果 结肠癌组织标本中miR-454表达69.86±7.29与V-ATPase mRNA表达5.42±0.41高于良性结肠病变组织10.74±2.03、2.79±0.38、癌旁正常结肠组织标本1.28±0.74、1.96±0.32,miR-132水平表达1.46±0.25低于良性结肠病变组织3.68±0.73、癌旁正常结肠组织标本4.59±1.24(P<0.05);中低...

Abstract:Objective To investigate the association of tissue microRNA (miR)-454, miR-132, and vesicular membrane-ATPase (V-ATPase) expression with the clinicopathological characteristics of patients with colon cancer.Methods Totally 96 colon cancer tissuespecimens, 96 paracancerous normal colon tissue specimens, and 82 benign colon lesion specimens, obtained from surgical resection atthe First Affiliated Hospital of Xinxiang Medical College from September 2011 to November 2014, were selected for the study. The ex.pressions of miR-454, miR-132 and V-ATPase mRNA in the specimens were measured by qRT-PCR, and the relationship between miR-454, miR-132 and V-ATPase mRNA expressions and general data and clinicopathological parameters of colon cancer patients was ana.lyzed. Patients were followed up for 5 years and the 3-year and 5-year survival rates were calculated of the patients with high or low miR-454, miR-132, and V-ATPase expressions.Results The expressions of miR-454 and V-ATPase mRNA were higher in colon cancer tis.sue specimens than in benign colon lesion tissues,and normal colon tissue specimens adjacent to cancer [miR-454: 69.86±7.29 vs. (10.74±2.03), (1.28±0.74); V-ATPase mRNA: (5.42±0.41) vs. (2.79±0.38), (1.96±0.32)], and miR-132 expression was lower than that of benign colon lesion tissues and normal colon tissue specimens adjacent to cancer (1.46±0.25 vs. 3.68±0.73, 4.59±1.24; P<0.05). The ex. pressions of miR-454 and V-ATPase mRNA were higher in patients with moderately low-differentiated, Ducks stage C to D, colon cancerwith lymph node metastasis than in patients with highly differentiated, Ducks stage A to B, and no lymph node metastasis, and miR-132 expression was lower than that in the patients with highly differentiated, Ducks stage A to B, and no lymph node metastasis (P<0.05). miR-454 and V-ATPase mRNA expressions were significantly negatively correlated with the degree of tissue differentiation and signifi.cantly positively correlated with Ducks staging and lymph node metastasis. miR-132 expression was significantly positively correlatedwith the degree of tissue differentiation and significantly negatively correlated with Ducks staging and lymph node metastasis (P<0.05). The 3-year (9.23%, 33.85%) and 5-year survival rates (44.00%, 28.00%) were lower in patients with high miR-454, V-ATPase mRNA expressions than in patients with low expressions (70.97%, 61.29%, 69.57%, 55.10%), and the 3-year, 5-year miR-132 high expression survival rates (70.59%, 58.82%) were higher than those of low expression patients (48.39%, 33.87%; P<0.05).Conclusions miR-454 and V-ATPase mRNA expressions showed a significant negative correlation with the degree of tissue differentiation and a significant posi.tive correlation with Ducks staging and lymph node metastasis. miR-132 expression showed significant positive correlation with the de.gree of tissue differentiation and significant negative correlation with Ducks staging and lymph node metastasis, which can provide an ob.jective basis for the characteristics of malignant biological behavior, pathological grade and prognosis of colon cancer.
