摘    要:目的探讨具有家族史的食管癌和正常食管组织内蛋白质组的表达差异,为寻找参与食管癌发生的相关蛋白奠定基础。方法采用双向凝胶电泳分离人食管癌和正常食管上皮组织的总蛋白图谱,经图像分析后,应用蛋白质谱仪对差异蛋白进行鉴定分析。结果食管癌组织中有(45±12)个蛋白为新增蛋白,同时相对正常食管上皮中有(9±3)个蛋白缺失。随机选择其中的18个位点进行质谱鉴定发现,9个蛋白质与肿瘤细胞的增殖、分化和肿瘤转移有关。其中肿瘤组织中特异表达的8个,缺失1个。结论建立了分辨率较高的食管癌组织双向电泳图谱,并识别鉴定出了一些与肿瘤相关的蛋白质,为进一步筛选食管癌特异性的分子标志物奠定了基础。

Abstract:Objective To analyze the difference in protein expression between normal esophageal tissue and esophageal cancer with family history,and then to lay the foundation for finding the specific proteins which may participate the carcinogenesis of esophagus.Methods The proteins were sperated from the two types tissue by two dimensional gel electrophoresis.The differential expression proteins were analyzed by using imaging master and identified by MALDI-TOF-MS.Results In the esophageal cancer tissue,the total of new protiens were (45 ± 12) ,and (9±3) proteins were absenced. 18 protein spots chosen randomly were identified by MALDI-TOF-MS,9 proteins were involved in the cell proliferation,differentiation and metastasis,8 of them were tissue-specific expressed in tumor, 1 was abscent. Conclusions The test establishs well high-resolution 2-DE profiles of esophageal cancer. Some proteins related to carcinogenesis of esophagus are sought. These results provide a basis for further study of the specific proteins markers of esophageal cancer.
