慢性支气管炎大鼠气道炎症与血清胆红素的动态观察及意义

摘    要:目的探讨脂多糖(LPS)致大鼠慢性支气管炎发生、发展中气道炎症与血清胆红素的变化特点。方法将96只雄性二级SD大鼠随机分为对照组和慢性支气管炎组(每组48只)。采用大鼠气管内注入脂多糖的方法制备慢性支气管炎大鼠模型;对照组气管内注入等量生理盐水。于术后不同时期动态观察:①血清总胆红素;②支气管肺泡灌洗液(BALF)中白细胞(WBC)、肿瘤坏死因子(TNF)-α、丙二醛(MDA)及超氧化物歧化酶(SOD)的变化。结果与对照组相比,慢性支气管炎组血清总胆红素水平在注药后降低,6 h时最低(P<0.05),以后缓慢回升;其BALF中WBC和TNF-α均以6 h最高(P<0.05),3周时次高(P<0.05),二者变化趋于一致;MDA在24 h最高(P<0.05),3周时次高(P<0.05);而SOD在6 h最低(P>0.05),1周时最高(P<0.05)。结论在慢性支气管炎气道炎症中,炎性细胞、细胞因子及氧化应激是造成气道损伤的重要原因,血清胆红素的降低是其原因之一。

Abstract:Objective To investigate the characteristic of the airway inflammatory reaction and the levels of total serum bilirubin in rat with chronic bronchitis during the development of the disease.Methods Ninety-six male Sprague-Dawley rats were divided into two groups(48 for each) at random,i.e.control group and chronic bronchitis model group.Rats' chronic bronchitis model was estabilished by intratracheal instillation of small-dosed lipopolysaccharide,as the chronic bronitis group.Rats were treated by intratracheal instillation of the same dose of normal saline in the control group.Levels of total serum bilirubin,total white blood cell(WBC) count,tumor necrosis factor(TNF)-α,malondialdehyde(MDA) and superoxide dismutase(SOD) of broncho-alveolar lavage fluid(BALF) were determined after operation.Results As compared with that of control group,the level of total serum bilirubin in chronic bronchitis group decreased after operation.Most significant decrease was found at the 6th h(P<0.05),then,it resumed gradually.Most significant increase in WBC and TNF-α in BALF was found at the 6th h(P<0.05),and more significant increase at the 3rd week(P<0.05).The same thing happened in MDA at the 24th h and the 3rd week,respectively.However,most significant decrease in SOD was found at the 6th h(P>0.05) and most significant increase at the 1st week(P<0.05).Conclusion Inflammatory cells,cytokines and oxidative stress are primary factors for damaging airway in the development of chronic bronchitis;total serum bilirubin's drop is also one of them.
