过期血液中血小板生长因子促细胞增殖作用的初步研究

摘    要:目的探讨过期血液中血小板生长因子含量及促细胞增殖作用。方法选取全血和浓缩血小板样品为研究对象,分别采用冻融法和CaCl2法处理保存d 1、5、10的血小板,应用ELISA试剂盒测定样品中PDGF-AB、TGF-β1的含量,应用CCK-8试剂盒测定样品的促细胞增殖作用;在全血保存d 1、10、21、35、40时取样,离心收集血浆,测定其血小板生长因子含量和促细胞增殖作用。结果 CaCl2法处理的d 1、5、10血小板样品中PDGF-AB含量分别为86.56±19.66、76.15±18.55、70.65±24.13 ng/ml,TGF-β1的含量分别为86.40±19.92、79.80±16.54、92.45±17.59 ng/ml;冻融法处理的d 1、5、10血小板样品中PDGF-AB含量分别为48.24±3.73、60.91±18.39、69.35±22.29 ng/ml,TGF-β1的含量分别为107.57±46.48 149.33±24.78 154.26±38.99 ng/ml,同一处理方法 d 1、5、10血小板样品中生长因子含量无明显差别(P>0.05);同一处理方法不同时间点血小板样品对细胞增殖作用无明显差别(P>0.05);冻融法处理后样品在浓度20%时促细胞增殖率明显下降;不同时间点全血血浆促细胞增殖作用趋势线相近,在样品浓度达到20%时促细胞增殖率均明显下降。结论过期血小板中血小板生长因子含量及促细胞增殖作用与新鲜血小板无显著差异,过期全血血浆中的血小板生长因子与新鲜全血相比也得到了相似的初步结果。

Abstract:Objective To investigate the proliferative activity of platelet growth factors in expired blood.Methods Platelets collected at the preservation of 1st,5th,10th day were processed by the two methods of freeze-thaw and CaCl2,the proliferative activity of the samples tested by the CCK-8 kit,and the content of PDGF-AB and TGF-β1 measured by the Elisa kits.The whole blood samples were collected at the preservation of 1st,10th,21st,35th,40th day,segregating the plasma at 1 000 g for 10 min.The proliferative activity of the plasma was measured by the CCK-8 Kit,and the growth factors were measured by the Elisa kits.Results The content of the PDGF-AB in the sample processed by CaCl2 at 1st,5th,10th day was(86.56±19.66),(76.15±18.55),(70.65±24.13)ng/ml and TGF-β1 was(86.40±19.92),(79.80±16.54),(92.45±17.59)ng/ml.The content of the PDGF-AB in the sample processed by freeze-thaw at 1st,5th,10th day was(48.24±3.73),(60.91±18.39),(69.35±22.29)ng/ml and TGF-β1 was(107.57±46.48),(149.33±24.78),(154.26±38.99)ng/ml,separately.The proliferative activity of platelet samples collected from different times have no significant difference(P>0.05).The proliferative activity of the platelet sample processed by free-thaw decreased at the 20% content,but it was not decrease at 20% content processed by CaCl2.The trend lines of the cell proliferation effected by the plasmas from different times were similar,but the activity still decreased at the 20% content.Conclusion Platelet growth factors in expired blood should be retain proliferative activity.
