CXCR3在结直肠癌组织中的表达及临床意义

目的 探讨CXCR3在结直肠癌组织中的表达及其临床意义.方法 应用SP免疫组织化学方法检测52例结直肠癌组织CXCR3的表达,Western印迹和体外迁移实验检测趋化因子受体CXCR3在大肠癌细胞株SW480、SW620、HT29细胞的表达及迁移能力.结果 CXCR3的阳性表达率为88%(46/52),CXCR3的高表达与结直肠癌患者的年龄、性别、结直肠癌组织类型、分化程度无关,而与临床转移(肝脏、淋巴结转移)有关(x2=12.1、21.5,P＜0.01).SW480、SW620及HT29的CXCR3表达均呈阳性,其中大肠癌细胞HT29和SW620 CXCR3高表达,大肠癌细胞SW480 CXCR3低表达.在体外迁移实验中,大肠癌细胞HT29和SW620发生体外迁移细胞明显增多(75±6比147±8、63±5比9l±6,P＜0.01),而大肠癌细胞SW480体外迁移无明显变化(47±4比45±3,P>0.05).结论 CXCR3上调表达与结直肠癌转移有关.

Expression of CXCR3 in colorectal cancer and its significance

Objective To investigate the expression and significance of CXCR3 in human colorectal cancer.Methods Immunohistochemistry was performed to detect the expression of CXCR3 on 52 colorectal cancer specimens,the CXCR3.expression of colorectal cancer cell line SW480,SW620 and HT29 was determined by Western blot,and migratory responses to stromal cell-derived factor 1 (SDF-1)were tested by cellular migration assays.Results In all 52 cases,the expression rate of CXCR4 was 88% (46/52).The CXCR3 expression was independent of age,gender,pathological type or histological grade.However,higher expression of CXCR3 tended to be detected in the tumor specimens with liver metastasis or lymph node metastasis (X2 = 12.1,21.5 ,P＜0.01 ).The CXCR3 expression in human eolorectal cancer cell lines varied from strong (HT29,SW620) to weak (SW480).In the in vitro cellular migration assays,a significant increase of cell migration was found in HT29 and SW620 (75±6 vs 147±8,63±5 vs 91±6,P＜0.01 ),but not in SW480 ( 47±4 vs 45±3,P＞0.05 ).Conclusion Up-regulated expression of CXCR3 was associated with coloreetal cancer metastasis.
