康欣胶囊对血管性痴呆模型大鼠皮质顶叶、海马CA1区神经生长因子表达的影响

摘    要:目的观察补肾健脾养血活血中药制剂康欣胶囊对血管性痴呆(VD)模型大鼠皮质顶叶、海马CA1区神经生长因子(NGF)的表达以及神经突触形态和数量的影响。方法光化学反应法制作VD模型大鼠,康欣胶囊高、中、低剂量及阳性西药喜得镇给药1个月,同时设立模型组和空白组,电镜观察各组大鼠皮质顶叶、海马CA1区神经突触形态,计算突触数密度,免疫组化染色观察皮质顶叶、海马CA1区NGF阳性细胞情况。结果电镜下,康欣胶囊各给药组及阳性西药组皮质顶叶、海马CA1区突触形态完好,数量正常,与空白组情况接近,优于模型组;两部位NGF阳性细胞显著多于模型组(P<0.01),与空白组接近。结论康欣胶囊可提高VD模型大鼠皮质顶叶、海马CA1区NGF的表达,保护突触形态和数量,从而保护缺血损伤后神经系统功能。

Abstract:Objective To observe the effect of Kangxin capsule on nerve growth factor (NGF) expression in cortical parietal lobe and hippocampus CA1 zone and quantity and structure of synapse of vascular dementia (VD) model rats. Methods The experimental VD rats, made by photochemically initiated thrombosis, were divided into six groups: model group, control group, high dosage group, middle dosage group, low dosage group and western medicine group. Then structure of synapse in the cortical parietal lobe and hippocampus CA1 zone of rats in every group was observed by electron microscope, synapse number density was calculated, and the NGF positive cells in cortical parietal lobe and hippocampus CA1 zone were observed with immunohistochemical quantitative analysis. Results Under electron microscope, each group treated with Kangxin capsule and western medicine group demonstrated a normal quantity and complete structure of synapse, was similar to that in control group, better than that in model group; and NGF positive cells in the cortical parietal lobe and hippocampus CA1 zone were significantly more than that of model group (P<0.01), similar to that of control group. Conclusions Kangxin capsule can induce the NGF expression in cortical parietal lobe and hippocampus CA1 zone of VD model rats, protect structure and quantity of synapse and promote neuroprotective effect after cerebral ischemia.
