人骨髓间充质干细胞体外优化条件的研究

目的通过比较5种不同体外培养人骨髓问充质干细胞（hMSCs）条件，找到一种能在质和量上均能满足需要的体外培养条件。方法使用Percoll-Paque液（1．073g／ml）分离人骨髓血，采用DMEM＋15%的小牛血清、人体白蛋白、人血清、脐带血清及MesenCult培养基，在相同培养条件下，动态观察hMSCs的生长状况、细胞数量变化，通过流式细胞仪检测细胞的纯化状况。结果5种培养条件在一定时相内均能获得较为纯化的hMSCs，但在生长速度和纯化率上存在一定的差异。结论Mesen Cult是一种较为优秀的hMSCs培养基．胎儿脐带血清培养也能在短期内获得足量纯化的hMSCs．人血清的使用还需要做进一步的研究。

Comparison of Several Culture Methods of Human Mesenchymal Stem Cells in Vitro

Objective Observe five culture methods of hMSCs.To find a better methods in vitro to satisfy in terms of quality and quantity.Methods Human bone marrow was separated with Percoll-Paque regent,cultured with DMEM+15%FCS,human blood serum,Alb,umbilical cord blood serum and MesenCult culture medium.In the same culture condition,we study hMSCs growing status,and cells quantity,and conduct detection with FCM(flow cytometry).Results All methods can obtain the purification hMSCs.But there was differentiation in growing velocity and purification rate.Conclusion MesenCult culture medium is better than the other culture methods in vitro.Umbilical cord blood serum can also obtain a great deal of purified hMSCs,the using of human blood plasma needs more investigation.
