内毒素条件下参与中性粒细胞-血管内皮细胞粘附过程的分子基础

摘    要:利用抗中性粒细胞(PMN)和血管内皮细胞(VEC)表面表达的粘附因子CD18、内皮细胞白细胞粘附分子-1(ELAM-1)及细胞间粘附分子-1(ICAM-1)单克隆抗体作为阻断因素,用细胞微管吸吮技术所测得的PMN-VEC间单细胞粘附力作为观察指标,观察了内毒素刺激所致PMN-VEC间粘附性增加过程中上述三种粘附分子各自的功能特征。结果发现:CD18的表达为PMN快相增加粘附力的主要分子基础,而ELAM-1、ICAM-1则为VEC慢相增加粘附力的主要分子基础。

Abstract:The monocellular adhesive ability of polymorphonuclear neutrophils(PMN)to vascular endothelial cells(VEC)was determined with the micromanipulation technique when the cells were treated with endotoxin and adhesion-interruptive factass of anti-CD18 monoclonal antibodies(McAb), anti-endothelialleucocytic adhesion molecule-1(ELAM-1)McAb and anti-intercellular adhesion molecule-1(ICAM-1)McAb and the functional characteristics of these 3 adhesion-interruptive molecules in the course of increased adhesion between PMN and VEC were observed.It was found that the main molecular basis of the rapid increase of PMN adhesive ability was CDI8 expression and that of the delayed increase of VEC adhesive ability was the expression of either ELAM-1 or ICAM-1.
