摘    要:目的:建立人血浆中百草枯浓度高效液相色谱检测方法,为百草枯中毒患者预后评估、治疗方案选择提供方法学支持。方法样品处理采用35%高氯酸处理。流动相为0.1mol · L -1磷酸缓冲液(含75 mmol · L -1庚烷磺酸钠)‐乙腈(88∶12,三乙胺调pH=3.0);流速为1.0 mL · min-1;柱温为28℃;检测波长为258 nm。结果所采用方法在0.2~500μg · mL -1范围内线性关系良好。平均绝对回收率为100.6%,RSD均小于3%;平均相对回收率为101.31%,RSD均小于6%。日内和日间RSD均小于6%,冻融及冻存后百草枯样品浓度测定结果差异无统计学意义。结论高效液相色谱检测方法快速,准确、血液中杂质无干扰,适用于百草枯中毒患者血药浓度的分析测定。

Abstract:Objective To establish an HPLC method for determination of paraquat in human plasma to provide methodology support for the assessment of prognosis and choice of therapy of patients with paraquat poisoning .Methods The plasma sam‐ples were deproteimized with 35% perchlorld acid .The mobile phase consisted of 0.1 mol · L -1 phosphate buffer (75 mmol · L -1 sodium heptanesulfonate)-acetonitrile (88:12) ,adjusted pH to 3.0 by triethylamine ,at a flowrate of 1.0 mL · min-1 ,and the detection occurred at 258 nm ,column temperature at 28℃ .Results A good linearity was obtained in the concentration range of 0.2~500 μg · mL -1 .The average absolute recovery was 100.6% ,RSD were less than 3% ,average relative recovery was 101.31% ,RSD less than 6% .The intra‐day and inter‐day RSD were less than 6% .After thawing and frozen ,concentration of paraquat showed no significant difference .Conclusion HPLC method was rapid ,accurate and without interference of impuri‐ties ,which could be applied in analysis and determination of plasma concentration for patients with paraquat poisoning .
