缬草单萜氧化物对单个兔心室肌细胞钠通道的影响

摘    要:利用全细胞膜片钳记录技术研究缬草单萜氧化物 (VMO)对兔单个心室肌细胞钠离子电流 (INa)的影响。结果 :30 μg/L和 6 0 μg/L的VMO使兔心室肌细胞INa峰值 (INamax)从 5 3.4 7± 5 .1 3pA/pF分别降至 4 0 .2 5± 4 .1 8pA/pF和 30 .89± 2 .95pA/pF(n =8,P <0 .0 5和P <0 .0 1 ) ,抑制率分别为 2 4 .7%和 4 1 .9% ;VMO使INa的电流 电压曲线上移 ,但不改变其激活电位、电位峰值和反转电位 ;VMO还减慢钠通道灭活后的恢复过程。结论 :VMO对INa具有浓度依赖性阻滞作用 ,这可能是其抗心律失常的重要机制之一。

Abstract:Whole cell patch clamp technique was used to investigate the effect of valeriana officinalis L extract(VMO) on sodium current(I Na ) in rabbit ventricular myocytes . Resultes : VMO at 30,60 μg/L decreased peak I Na (I Namax ) from 53.47±5.13 pA/pF to 40.25±4.18 pA/pF and 30.89±2.95 pA/pF( n=8,P<0.05 and P<0.01).VMO upshifted the I Na I V curves but didn′t change their active, peak and reverse potentials; VMO at 60 μg/L turned the steady state inactivation curve to right. Conclusion: VMO blocks I Na in a concentration dependent manner and probably inhibits I Na in its inactive state, which may contribute to its antiarrhythmic effect.
