摘    要:目的 探讨高胆固醇饮食对血清及内脏脂肪组织脂肪因子(瘦素、抵抗素、内脂素、脂联素、磷脂酰基醇蛋白聚糖-4)水平的影响.方法 SD大鼠60只随机分为普通饮食组(NC组,100%标准饮食)和高胆固醇饮食组(HC组,高胆固醇饮食),每组30只,喂养至第0、4、12、18周留取血样及内脏脂肪组织.检测空腹血糖、空腹胰岛素水平,计算胰岛素抵抗指数.酶联免疫吸附法检测血清和内脏脂肪组织中脂肪因子的水平.结果 与NC组相比,HC组大鼠喂养至第18周血清内脂素、脂联素显著升高(P<0.05),内脏脂肪组织中瘦素、内脂素、脂联素、磷脂酰基醇蛋白聚糖-4显著升高(P<0.05),并出现胰岛素抵抗.结论 长期过量胆固醇摄入导致大鼠脂肪合成及分泌脂肪因子功能紊乱,可能与胰岛素抵抗发生密切相关.

Abstract:Objective To investigate the effects of high cholesterol diet on adipokines (leptin, adiponectin, resistin, visfatin and glypican4) levels in serum and visceral adipose tissue (VAT). Methods A total of 60 SD rats were randomly divided into normal diet group (NC, standard diet, n=30) and high cholesterol diet group (HC, high cholesterol diet, n=30). Blood samples and VAT were collected at week 0, 4, 12 and 18, respectively. The levels of fasting blood glucose and fasting serum insulin were measured, and insulin resistance values were calculated. Adipokines in serum and VAT were detected with enzyme-linked immunosorbent assays. Results At week 18, the serum visfatin and adiponectin in the HC group were significantly increased compared with those in the NC group (P<0.05); levels of leptin, visfatin, adiponectin, and glypican4 in VAT were significantly higher in the HC group than in the NC group (P<0.05); insulin resistance was also observed in HC group. Conclusion Long-term excessive cholesterol intake induces disorder of adipokine synthesis and secretion, and such disorder may be correlated to insulin resistance.
