摘 要: | 目的:观察当归拈痛汤对高尿酸血症性肾病模型小鼠的疗效并探索其作用机制。方法:将72只小鼠分为模型组,空白对照组,当归拈痛汤高、中、低剂量组[38、19、9.5g/(kg·d)],苯溴马隆组。先采用腺嘌呤(100mg/kg)联合乙胺丁醇(250mg/kg)法建立高尿酸血症性肾病小鼠模型,再连续给药14d。实验结束后将小鼠眼球取血,分光光度法检测血清中一氧化氮(NO)、血尿素氮(BUN),酶比色法检测血清中血尿酸(BUA)、肌酐(CRE),ELISA法检测血清中内皮型一氧化氮合酶(eNOS)、内皮素-1(ET-1)水平。结果:当归拈痛汤可以显著降低BUA、BUN、CRE含量,升高eNOS水平,提高NO含量,降低ET-1水平,以当归拈痛汤高剂量组最为明显(P<0.01)。结论:当归拈痛汤可以通过降低BUA含量而升高血中eNOS、NO水平,进而降低ET-1水平,达到对肾脏组织内皮细胞及肾小球血管的保护,从而降低BUN、CRE含量,发挥对本病的治疗作用。本方治疗高尿酸血症性肾病的其他作用机制有待进一步探索。

Study on the mechanism of Danggui Niantong decoction for hyperuricemic nephropathy mice |

Abstract: | Objective:To observe the curative effect of Danggui Niantong decoction against hyperuricemic nephropathy and to explore the mechanism. Methods:The animals were divided into normal group,model group,positive drug Benzbromarone,the low dose of Danggui Niantong decoction group(9.5 g/kg),the middle dose(19 g/kg) and high dose(38 g/kg).Animal model was made by Adenine(100 mg/kg) and Ethylamine butanol(250 mg/kg).Then,the drug with different doses was given to different groups for 14 days. At the end of the experiment,the blood was collected from each group. Then,the serum level of NO,BUN,BUA,CRE,eNOS,ET-1 were measured. Results:Danggui Niantong decoction significantly reduced the level of ET-1,BUN,BUA,CRE,and increased the expression of eNOS and NO. Conclusion:Danggui Niantong decoction can reduce BUA production and increase serum eNOS,NO. Danggui Niantong decoction play a protective role on the kidney by reducing the level of ET-1.In the meanwhile,Danggui Niantong decoction reduced the level of BUN and CRE in serum. The other mechanism of the treatment of hyperuricemia nephropathy needs to be further explored. |
