摘    要:目的:观察参鱼化痰口服液对慢性阻塞性肺疾病(COPD)大鼠气道上皮中杯状细胞和粘蛋白MUC5AC表达的影响,探讨参鱼化痰口服液干预COPD大鼠气道黏液高分泌的可能机制。方法:清洁级SD大鼠随机分为空白组、模型组和中药组(参鱼化痰口服液组),每组12只,按照文献方法建立大鼠COPD模型。支气管肺组织标本中杯状细胞采用阿尔辛蓝过碘酸希夫(ABPAS)染色法检测,以阳染率表示,MUC5AC的表达采用免疫组化法检测。结果:与空白组比较,模型组气道上皮中杯状细胞数显著增加,差异有统计学意义(P<0.01),MUC5AC的表达显著升高,差异有统计学意义(P<0.01)。与模型组比较,中药组杯状细胞数明显减少,差异有统计学意义(P<0.05),MUC5AC的表达明显降低,差异有统计学意义(P<0.05)。结论:参鱼化痰口服液可抑制COPD大鼠气道上皮中杯状细胞增生,减少黏蛋白表达,减轻气道黏液高分泌,其机制可能是通过EGFR-MUC5AC信号通路来实现。

Based on EGFR-MUC5AC signaling pathway to explore the mechanism of Shenyu Huatan oral liquid in intervention of airway mucus high secretion in COPD rats

Abstract:Objective:To observe the effect of Shenyu Huatan oral liquid on the expression of goblet cells and mucin MUC5AC in airway epithelium of COPD rats,and to explore the possible mechanism of Shenyu Huatan oral liquid in interfering with airway mucus hypersecretion in COPD rats. Methods:SD rats were randomly divided into blank group,model group and Chinese medicine group (Shenyu Huatan oral liquid group),12 rats in each group. The rats COPD model were established according to the literature method. In the bronchopulmonary tissue samples,the goblet cells were detected by Alcian blue periodic acid Schiff (ABPAS) staining,and the expression of MUC5AC was detected by immunohistochemistry. Results:Compared with the blank group,the number of goblet cells in the airway epithelium of the model group was significantly increased ( P <0.01),and the expression of MUC5AC was significantly increased ( P <0.01). Compared with the model group,the number of goblet cells in the Chinese medicine group was significantly reduced ( P <0.05),and the expression of MUC5AC was significantly decreased ( P <0.05). Conclusion:Shenyu Huatan oral liquid can inhibit goblet cell hyperplasia in airway epithelium of COPD rats,reduce mucin expression and reduce airway mucus hypersecretion. The mechanism may be achieved by EGFR-MUC5AC signaling pathway.
