摘    要:目的:建立异种肝细胞皮下移植模型,并探讨用以研究异种移植细胞性排斥反应的可能性。方法:用原位肝脏胶原酶循环灌注法分离大鼠肝细胞,分离出的肝细胞存活率达95%以上,分别注射于皮下及脾脏,于移植后不同时间点取材进行组织形态及功能观察,HE染色为主,结合G-6P、SDH等组织化学方法观察移植肝细胞的存活状态。结果:在术后3-28天期间,脾脏组与皮下组存活率无显著性差异(P>0.05),仅在60天时相点脾脏组存活率(75%)大于皮下组(37.5%);且术后4-7天开始发生细胞性排斥反应。结论:认为肝细胞皮下移植可作为研究异种移植细胞性排斥反应的模型。

Abstract:Objective :This study is to transplant hepatocyte subcutaneosly and to see the possibility of eliminating CMXR by using this new model. Methods Preparation of isolated hepatocytes was performed fundamentally according to the collagenase digestive method of Berry and Friend using a recirculating perfusion apparatus. Hepatocellular transplantation was carried out by direct injection of the hepatocyte suspension into the splenie parenchyma or injecting subcutaneously. Recipients were killed, and their spleen and skin specimens were fixed in 10% formalin and stained with hematoxylin and eosin and for PAS. For G - 6 - P and SDH stain, fresh thin frozen sections were used. Results The results revealed that no significant difference of the xenograft survival was found between splenic hepatocyte and subcutaneous hepatocyte within one month. It is always believed that the CMXR happen from four to seven days after transplantation. Conclusion So the subcutaneous hepatocellular transplantation can be used as the model to appreciate CMXR.
