红细胞生成素测定方法的建立

摘    要:建立简便、经济、快速、敏感的红细胞生成素测定方法。方法通过测定盐酸苯肼处理小鼠脾细胞对3H-TdR掺入率的变化,间接反映样品中红细胞生成素的浓度。结果上述脾细胞对3H-TdR的掺入率与培养液中红细胞生成素的浓度呈正相关,敏感性高,稳定性强。结论RPMI1640培养液适合上述脾细胞生长。该红细胞生成素的测定方法可用于临床,特别适用于大样本的检测。

Abstract:Obiective To establish the microassay forerythropoietin, which with the characteristics ofsimplicity, economy, rapidity and sensibility.Methods Based on 3H-TdR incorporation. into spleen cells treated with phenylhydrazinehydrochloride from mice, the concentration oferythropoietin may be indiretly refleted by changeof 3H-TdR incorporation rates:Results The incorporation of 3H-TdR intothe spleen cells was positively related to the dosageof erythropoietin added in the media.Conclusions The medium of RPMI 1640 fitsthe growth of the spleen cells.The microassay foreryhropoietin can be practically used, especiallyfor more blood samples.
