柴胡皂苷d在小鼠体内的类雌激素样作用

摘    要:目的:探讨柴胡皂苷d在小鼠体内是否具有类雌激素样作用。方法:40只雌性BALB/c小鼠随机分为假手术组、模型组、雌二醇组和大剂量(50mg/kg)、小剂量(5mg/kg)柴胡皂苷d组,每组8只。除假手术组外,各组小鼠行双侧卵巢切除术造模。卵巢切除术后第10天,予相应药物腹腔注射治疗2周,观察小鼠阴道涂片细胞角化情况和体质量变化;末次给药12h后眼眶取血,放射免疫法检测小鼠血清雌二醇水平;剥离子宫、肾上腺,称取质量,计算子宫和肾上腺指数。结果:大、小剂量柴胡皂苷d组上皮细胞角化率高于模型组(P〈0.05),低于雌二醇组(P〈0.05);大、小剂量柴胡皂苷d组血清雌二醇水平高于模型组(P〈0.05),小剂量柴胡皂苷d组低于雌二醇组(P〈0.05);大、小剂量柴胡皂苷d组子宫指数和肾上腺指数高于模型组(P〈0.05),低于雌二醇组(P〈0.05);大剂量柴胡皂苷d组和雌二醇组体质量低于模型组(P〈0.05)。结论:柴胡皂苷d在体内具有弱雌激素样作用,可能是一种潜在的植物雌激素。

Abstract:Objective: TO investigate whether saikosaponin-d (SSd) had estrogen-like effects in mice. Methods: Forty female BALB/c mice were randomly divided Into sham.operated group, untreated group. estradiol (E2) group, low-dose SSd (5 mg/kg) group and high-dose SSd (50 mg/kg) group, with 8 mice in each group. Except the sham-operated group, bilateral ovariectomy was performed in the mice under anesthesia with 1. 25 % pentobarbital (40mg/kg). Ten days after the operation, the mice were administered with corresponding medicine by intraperitoneal injection once each day. Two weeks later, the rates of epithelial cornified cells of vagina in the mice were examined, and the body weights, uterus weights and adrenal weights were also measured. Twelve hours after the final administration, serum estradiol levels were tested by radioimmunoassay. Resulls: The rates of epithelial cornified cells of vagina in mice in the high-dose SSd group and the low-dose SSd group were higher than that in the untreated group (P〈0. 05), but were lower than that in the E2 group (P〈0. 057. Serum E2 levels of mice in the SSd groups were higher than thai in the untreated group (P〈0.05), but the serum E: level of the low-dose SSd group was lower than that in the E: group (P〈0. 05), Uterus and adrenal indexes of mice in the SSd groups were htgher than that in the untreated group ( P〈 0.05), however, they were lower than that in the E, group (P〈0. 05). The body weights of the mice in the high-dose SSd group and the E2 group were lower than that in the untreated group (P〈0. 05). Conclusion: SSd has weak estrogen-like effects in mice and may be a potential phytoestrogen.
