摘    要:目的 建立一种高效率的原代小胶质细胞体外分离纯化方法.方法 以McCarthy等经典的小胶质细胞原代培养培养方法为基础,并进行改进,使用低浓度胰酶消化法帮助小胶质细胞从混合培养的胶质细胞分离下来,用OX42抗体免疫组化染色鉴定.结果 获得了高纯度的小胶质细胞.结论 低浓度胰酶消化法分离小胶质细胞与经典的振摇分离法所获得的纯度相当,但提高了细胞的分离效率.

Abstract:Objective To establish a method of improved efficient primary mouse microglia isolation and its purification.Methods Primary microglia culture was modified based on the classic microglia culture method of McCarthy's. Trypsinization was employed with 0.05% trypsin for the isolation of the microglia from mixed cultured cells. Microglial cells were identified by immunohistochemistry using mouse anti rat CD11b monoclonal antibody(OX42).Results High purity of Primary microglial cell was obtained.Conclusion The purity of microglia by the method of mild trypsinization was equal to that of McCarthy's. but the efficiency of microglia isolation is improved.
