甲基苯丙胺对大鼠小脑神经瘤细胞的毒性作用

目的 :探讨甲基苯丙胺对大鼠小脑神经瘤细胞凋亡的诱导作用。方法 :应用MTT法 ,乳酸脱氢酶 (LDH)法检测细胞增殖和毒性作用 ,观察甲基苯丙胺的神经毒性 ;应用DNA琼脂糖凝胶电泳和流式细胞仪观察甲基苯丙胺致神经细胞凋亡作用。结果 :( 1)甲基苯丙胺 ( 5 0 - 5 0 0 μmol·L- 1 )呈剂量依赖性抑制R2细胞增殖 ;( 2 )甲基苯丙胺 ( 2 5 0 ,5 0 0 μmol·L- 1 )作用 4 8h ,增加R2细胞LDH的漏出 ,分别比对照组高 6 4 %和 10 8% (P <0 0 5 ) ;( 3)细胞DNA琼脂糖凝胶电泳呈现典型的DNA梯状条带 ;( 4 )应用流式细胞仪检测 ,在G1 峰前出现一个“G1 亚峰” ,凋亡细胞所占比例为 2 6 8%。结论 :甲基苯丙胺对神经细胞有毒性作用 ,可促进神经细胞的凋亡

A STUDY ON TOXICITY OF CEREBELLUM NEURAL CELLS INDUCED BY METHAMPHETAMINE

Objective : To investigate the effect of methamphetamine-induced apoptosis of the rat cerebellum cell line R2. Methods : The neurotoxicity of methamphetamine to R2 cells was detected by MTT reduction assay and LDH leakage assay, and the apoptosis induced by methamphetamine in R2 cells was observed with DNA gel electrophoresis and flow cytometer. Results : (1) Methamphetamine (50-500 μmol·L -1 ) dose-dependently inhibited cell proliferation; (2) in R2 cells treated with methamphetamine (250, 500 μmol·L -1 ) for 48 h, LDH leakage ratio was 6 4%, 10 8% higher than that of the control group( P <0 05); (3) DNA gel electrophoresis revealed DNA ladder; (4) methamphetamine (500 μmol·L -1 ) elicited the apoptotic DNA peak of the cells (i.e. sub-G 1-peak) through flow cytometry. The apoptotic cell ratio was 26 8%. Conclusion : Methamphetamine has neurotoxic effects on R2 cells and can induce R2 cells apoptosis.
