Toll样受体-2对角质形成细胞分泌IL-8的影响

目的:观察白念珠菌刺激人角质形成细胞后IL-8的变化以及Toll样受体2(Toll-like receptor2,TLR-2)对IL-8分泌的影响,探讨角质形成细胞在皮肤免疫系统中的作用。方法:白念珠菌分别与抗TLR-2抗体预处理前后的角质形成细胞孵育,ELISA方法检测不同时间培养上清液IL-8的水平。结果:白念珠菌可引起IL-8明显升高(P<0.01),3h开始,至24h达到高峰;中和TLR-2受体后,虽然引起IL-8升高,但无统计学意义(P>0.05)。结论:白念珠菌可以刺激角质形成细胞分泌IL-8,TLR-2在角质形成细胞IL-8表达中的作用尚难确定。

Effects of Toll-like Receptor-2 on Interleukin-8 Secretion of Keratinocytes

Objective To observe the change of Interleukin-8 （IL-8） in keratinocytes induced with Candida Albicans and the effect of Toll-like receptor-2（TLR-2） on IL-8 secretion,to explore the role of keratinocytes in skin immune system. Methods Candida Albicans were cultured on keratinocytes treated before and after anti-TLR-2 monoantibody（mAb） ,respectively. The levels of IL-8 in culture supernatant were determined with ELISA at different time points. ReSultS The IL-8 levels were significantly increased after induced with Candida Albicans （ P 〈 0.01 ）, the significant increase was observed at 3 h and reached the peak at 24 h; the increase of IL-8 levels had no significant difference after TLR-2 was neutralized with anti-TLR-2 mAb（P 〉0.05）. Conclusion The secretion of IL-8 in keratinocytes could be induced with Candida albicans, the function of TLR-2 is uncertain in expression of IL-8 in keratinocytes.
