摘    要:黏液是一种黏弹性和可黏着的凝胶,用来保护呼吸道、胃肠道、生殖道、眼睛和其他黏膜表面免受外来微粒及病原体等有害物质的侵入,同时也能阻碍和限制药物微粒载体在其中的有效扩散及吸收,因此,黏膜给药的生物利用度往往较低。本文简述了黏液的生理学特性和阻滞微粒渗透的机制,介绍了一种新型的黏膜药物载体——穿透黏液粒子(MPP),以及纳米粒子在黏液等复杂的生物屏障中传递方式及扩散速率的定量方法——多粒子追踪(MPT)技术。

Abstract:Mucus is a type of viscoelastic and adhesive gel, which protects respiratory tracts, gastrointestinal tracts, reproductive tracts, eyes and other mucosa surfaces from the invasion of external microparticles and pathogens. The mucus, however, also inhibits the drug delivery and absorption through the mucosa surfaces, which reduces the bioavailability of the drug delivery system. This article introduces an innovative sustained and targeting mucosa drug delivery system, mucus-penetrating particles (MPP) and describes a characterization protocol to quantify the diffusion of nanoparticles through complicated biological barriers (such as cell cytoplasm and mucus), which is multiple-particle tracking (MPT).
