荧光与亚甲蓝联用对血浆中病毒灭活的研究

摘    要:以细胞感染试验、凝血法、交叉免疫电泳等技术 ,观察荧光与亚甲蓝联用对血浆中水泡性口炎病毒 (VSV)和辛德比斯病毒 (SV)的灭活效果及血浆蛋白的变化。结果 ,向盛装在聚氯乙烯袋内的血浆中加 1μmol/L亚甲蓝并经荧光 (30 0 0 0Lux)照射 30min ,可将血浆中滴度 >7TCID50 (lg)的VSV和SV灭活。消毒后 ,血浆中的Ⅷ :C、Ⅸ :C、纤维蛋白原的回收率 >75% ,白蛋白、球蛋白的回收率 >85% ,蛋白免疫原性无变化

Abstract:Techniques such as cell infection test, blood coagulation method and crossed immunoelectrophoresis were used to observed the efficacy of combination of fluorescence and methylene blue in inactivating vesicular stomatitis virus (VSV) and Sindbis Virus in plasma and the changes in plasma protein The results showed that addition of 1 μmol/L methylene blue to the plasma in polyvinyl chloride bag which was also exposed to fluorescence (30000 Lux) irradiation for 30 min could inactivate VSV and SV with titers >7 TCID 50 (lg) in plasma After disinfection, the recovery rates of FⅧ:C, FⅨ:C and fibrinogen were >75% and the recovery rates of albumin, globulin (IgG, IgA, IgM) were >85%, while no change in protein immunogenicity was observed
