宫颈鳞癌组织和细胞系中P120ctn及E-cad的表达及生物学意义

摘    要:目的:探讨P120连环蛋白(P120catenin,P120ctn)及E-钙粘蛋白(E-cadher-in,E-cad)在宫颈鳞癌组织和宫颈鳞癌细胞系(Caski、Siha)的表达、分布及其生物学意义。方法:免疫组织化学法检测宫颈鳞癌组织,免疫细胞化学法及RT-PCR检测宫颈鳞癌细胞系中P120ctn及E-cad的表达与分布。结果:(1)正常宫颈组织中P120ctn及E-cad在胞膜,胞浆少见表达;宫颈鳞癌中P120ctn及E-cad胞膜表达降低,出现胞浆表达,P120ctn及E-cad的异常表达与癌细胞分化程度、淋巴结转移有关,与年龄、临床分期无关;(2)正常对照细胞即人脐静脉内皮细胞系(HECV)中P120ctn大部分在胞膜表达,Caski、Siha细胞系中P120ctn胞膜表达量减少,胞浆表达量增加;E-cad在HECV中胞膜胞浆表达较少,Caski、Siha中胞浆表达较少,胞膜表达更少;(3)HECV及Caski、Siha细胞系中都有P120ctn与E-cad mRNA表达,P120ctn mRNA在HECV、Caski、Siha细胞系表达量无显著性差异(P>0.05),E-cad mRNA在Caski、Siha细胞系中表达量低于HECV(P<0.05)。结论:P120ctn及E-cad的异常表达与宫颈鳞癌发生发展密切相关,其异常表达与肿瘤细胞分化程度、淋巴结转移有关,两者异常变化可作为宫颈癌变的参考依据。

Abstract:Objective:To investigate the expression distribution and significance of P120-catenin(P120ctn) and E-cadherin (E-cad ) in samples of squamous cervical carcinoma(SCC) and SCC cell lines(Caski,Siha ).Methods:Expression and distribution of P120ctn and E-cad were examined in SCC tissue by immunohistochemical staining (SP method) and in Caski and Siha by immuocytochemistry(ICC) and RT-PCR.Results:(1) In the normal cervical epithelia tissue,P120ctn and E-cad were stained at the membrane and low expression at the cytoplasm.In SCC,P120ctn and E-cad were found at the cytoplasm,and reduced expression at membrane.Abnormal expression of P120ctn and E-cad were significantly correlate with tumor differentiation status and lymph node metastasis but not with clinical stage and age;(2) In the cell lines,P120ctn were found at the membrane in HECV,but the P120ctn was mainly expressed at the cytoplasm in Caski and Siha;E-cad expression was low at the membrane in HECV,but the E-cad expression was found even less in both of the cytoplasm and membrane in Caski and Siha;(3) mRNA expression of P120ctn and E-cad were found in the cell lines (HECV,Caski and Siha).P120ctn mRNA was not different between normal and cancer cell lines,but E-cad mRNA expression was much lower in Caski and Siha than that of HECV.Conclusions:Abnormal expression of P120ctn and E-cad are significantly correlated with carcinogenesis and development of SCC.The abnormal expressions of the two proteins above will be the indicators for the diagnosis of cervical carcinoma.
