碱性成纤维细胞生长因子对癫痫大鼠海马神经元Caspase-3表达的影响

摘    要:目的研究碱性成纤维细胞生长因子(bFGF)对癫痫大鼠海马神经元的神经保护作用及可能的机制。方法戊四氮诱发大鼠癫痫发作后,腹腔分别注射生理盐水(B组)和bFGF(C组),与空白对照组(A组)按一定时段断头取脑,进行HE染色和Caspase-3免疫组织化学染色。观察各组大鼠海马神经元的损伤以及Caspase-3的表达。结果HE染色:B组神经元形态散乱,大量神经元凋亡、坏死;C组可见散在的神经元凋亡,变性坏死少见。Caspase-3免疫组化染色:B组6 h可见海马CA1、CA3和齿状回门区部分细胞的胞浆和核内有棕黄色沉淀物,24 h阳性细胞数及着色深度达到高峰,持续至72 h,120h后表达减弱。C组阳性细胞数及染色度与B组有相似的时间依赖性,但总体表达较B组弱,组间各时间点中12~72 h的差异均有统计学意义(P<0.01),6 h及120 h的差异则无统计学意义(P>0.05)。结论bFGF能显著减轻癫痫所致的大鼠海马神经元损伤。

Abstract:Objective To investigate the neuron-protective function and the possible mechanism of basic fibroblast growth factor(bFGF) to hippocampal neurons of epileptic rat.Methods Epileptic seizure in rat model was induced by pentylenetetrazol and followed by normal saline(group B) or bFGF-abdomen-injection(group C),normal rats without epilepsy were taken as blank control group(group A).HE staining and immunohistochemistry technique were carried out to observe the alteration of neurons and the expressions of Caspase-3.Results HE staining:neuronal degeneration was observed in area CA1,CA3 and dentate hilus in both Group B and Group C,while cell degeneration and apoptosis in Group C was less than those in Group B.Caspase-3 immunohistochemistry staining showed neuron apoptosis in Group C was less than those in Group B at different time points(P<0.01).Conclusion The bFGF could reduce the hippocampal neuronal damage in epileptic rats significantly,which suggested that bFGF could regulate the expression of Caspase-3 gene and sequentially protect neurons.
