| 摘 要： | 目的检测单纯疱疹病毒性脑炎(HSE)小鼠脑组织中S100BmRNA的含量,初步探讨S100BmRNA对HSE小鼠脑损害的评估价值。方法使用已制造成功的HSE小鼠模型48只,分为HSE组、正常组、假手术组,分别提取小鼠脑组织总RNA,反转录为cDNA,采用RT-qPCR检测S100BmRNA含量,并对S100BmRNA进行测序。结果 HSE组S100B含量显著高于假手术及正常组(P<0.01);S100B基因测序结果在BLAST中进行比对,显示同源性100%证实合成S100B基因片段的正确。结论 S100B在HSE、正常、假手术组均有表达,但其表达的程度并不一致,提示S100B可能为HSE小鼠脑损害的标志物,其含量的高低与损害程度有关。

| Abstract: | Objective To detect levels of S100B in brain tissue of mice with herpes simplex virus encephalitis(HSE) and to explore the value of S100BmRNA on preliminary study on assessment of brain damage in HSE mice.Methods 48 HSE mice were divided into HSE,normal,sham-operated group.Total RNA were extracted from mouse brain and reverse transcribed into cDNA.S100B mRNA levels were detected using RT-qPCR.Results S100B in HSE group were significantly higher than those in the sham and normal group(P<0.01).S100B gene sequencing showed 100% homology S100B synthesis.Conclusion S-100B were successfully expressed in the HSE,the normal,sham group.HSE may prompt S100B marker of brain damage in mice and the levels are correlated with the degree of damage. |
